Cell Line Maintenance Protocols for Leukemia Research

The human leukemia cell lines K562 [28 (link)], HEL [29 (link)], MEG–01 [30 (link)], CMK-11-5 [31 (link)], HL60 [32 (link)], NB4 [33 (link)], Jurkat [34 (link)], Reh [35 (link)], and THP–1 [36 (link)] were obtained from the Japanese Collection of Research Bioresources Cell Bank and maintained in Roswell Park Memorial Institute 1640 medium (Life Technologies) supplemented with 10% fetal bovine serum (FBS) at 37°C in a humidified atmosphere of 5% CO2, unless otherwise indicated. The erythropoietin-dependent erythroid cell line UT–7/Epo was kindly provided by Dr. Norio Komatsu and maintained in the same medium supplemented with 10% FBS and 1 U/ml recombinant human erythropoietin (Kyowa Hakko Kirin, Tokyo, Japan) [37 (link)]. The Chinese hamster ovary cell line CHO-TRVb, which lacks functional TFR1, was kindly provided by Dr. Timothy McGraw and maintained in Ham’s F12 nutrient mixture (Life Technologies) supplemented with 5% FBS [38 (link)].

Culture of Leukemia Cell Lines

Human leukemia cell lines U937, Molt-4, and Jurkat cells were obtained from the Japanese Collection of Research Bioresources (JCRB) Cell Bank. Cells were cultured in RPMI 1640 medium supplemented with 10% heat-inactivated fetal bovine serum (FBS) (JRH Biosciences, Corston, UK) at 37.0°C in humidified air with 5% CO2. Viability of untreated control cells was over 95% in all experiments.

Cell Line Characterization and Reagents

HEK 293, HeLa, Jurkat, and NIH-3T3 cells were obtained from JCRB Cell Bank (Osaka, Japan). MC 38 and K562 cells were obtained from the Cell Resource Center for Biomedical Research, Institute of Development, Aging and Cancer, Tohoku University. PEG-CLS was purchased from Biochempeg (Watertown, MA, USA). BSA-CLS was created by bioconjugation of BSA (Merck, Tokyo, Japan) and NHS-PEG1K-cholesterol (Nanocs, New York, NY, USA). Gelatin-CLS was synthesized as previously described 11 . Calcein-AM and Cell Counting Kit-8 were purchased from Dojindo (Kumamoto, Japan).
