Quantifying Cellular Apoptosis via Flow Cytometry

Cell apoptosis was assessed by measuring the membrane redistribution of phosphatidylserine using PE Annexin V Apoptosis Detection Kit l (BD Biosciences, Franklin Lakes, NJ, USA), according to the manufacturer’s instructions. Briefly, cells (4×106/well) were seeded into a T25 cell culture bottle. After transfection, the cells were cultured for 36 h. Subsequently, the cells were in the logarithmic phase, washed twice with cold PBS, resuspended in 1×Binding Buffer at a concentration of 1×106 cells/mL, and then 100 µL of the solution (1×105 cells) was transferred to a 2 mL culture tube. Finally, cells were stained with 5 µL of PE and 5 µL of 7-AAD, gently vortexed and incubated for 15 min at RT in the dark. Then, the cells were filtered with a filter screen and analysed using the BD FACSCalibur flow cytometer (BD Biosciences, Franklin Lakes, NJ, USA). The cells in Q2 (late stage apoptosis) and Q4 (early stage apoptosis) area were all calculated as apoptotic cells.

Apoptosis Analysis by Flow Cytometry

Flow cytometry analysis was done using PE Annexin V apoptosis detection kit l (BD Biosciences, Haryana, HR, USA), following manufacturer’s instructions. Briefly, cells were seeded into 12-well plates at 2.5 × 105 per well. After treatment, trypsinized cells were resuspended in 500 µL of Binding Buffer 1X and subsequently stained with annexin V-PE and 7AAD. Stained cells were evaluated with the BD Accuri C6 flow cytometer (BD Biosciences, Haryana, HR, USA) and analyzed using the FlowJo software (BD Biosciences, Haryana, HR, USA; version 10.8.1 for Windows).
