Streptozotocin-Induced Diabetic Mouse Model

The male C57BL/6 mice (8 weeks old) were purchased from Guangzhou Medical University. Procedures were approved by the Institutional Animal Care and Use Committee of Guangzhou Medical University (No. SYXK2016-0168). Eight week old mice were injected with 50mg/kg streptozotocin (STZ, Sigma) to induce diabetes for 5 consecutive days. The mice were randomly divided into four groups and were intraperitoneally injected PBS or calpeptin (calpain inhibitor) (30 mg/kg/day) diluted in DMSO in PBS every other day. The mice in the AgomiR group were administered with Agomir-223 200 μl at a concentration of 20 mM via tail vein injection. The calpeptin ware purchased from the SelleckChemicals Co, while the AgomiRs and miRNA mimic from Shanghai GenePharma Co.

TET2 Knockdown in SH-SY5Y Neuroblastoma Cells

SH-SY5Y neuroblastoma cells were cultured in Dulbecco's modified eagle's medium (DMEM, Gibco) containing 10% fetal calf serum (Gibco, Brazil) and grown in a CO2 incubator maintained at atmospheric oxygen levels and 5% CO2. Dissolved MPP+ (Sigma Aldrich) in phosphate buffered saline (PBS) to a storage concentration of 125 mM. Resveratrol (Sigma) was prepared in dissolved dimethyl sulfoxide (DMSO) at a stock of 25 mM. EX-527, SRT1720, CHX, MG-132, Calpeptin, Z-VAD, and Bobcat339 (Selleckchem) stocks were dissolved in DMSO at the concentrations of 10 mM,10 mM, 1 mg/ml, 10 mM, 10 mM, 50 mM, and 3 mM, respectively. They were both stored at −20°C. Resveratrol, EX-527, and SRT1720 were pretreated cells 12 h before specific administration.
TET2 knockdown by lentiviral delivery of a short hairpin RNA (shRNA) was achieved via lentiviruses expressing the green fluorescent protein (GFP) and shRNA-TET2. shRNA-TET2 containing human shTET2a and shTET2b. Sequences for human shTET2a and shTET2b were (5′-GGGTAAGCCAAGAAAGAAA-3′) and (5′-AAACAAAGAGCAAGAGATT-3′), respectively.

Multiple Myeloma Cell Line Maintenance

U266 (ATCC Cat# TIB-196) and MM.1s (ATCC Cat# CRL-2974) multiple myeloma (MM) cells, and HS-5 (ATCC Cat# CRL-11882) bone marrow stroma cells were purchased from American Type Culture Collection (Manassas, VA, USA), they were maintained in RPMI-1640 medium with L-glutamine (Gibco; Life Technologies, Waltham, MA, USA) with 10% Fetal Bovine Serum (FBS) and 1% penicillin/streptomycin. The cell lines were validated by short tandem repeat (STR) and they were also tested for mycoplasma every six months. MTI-101 was synthesized by Bachem (San Diego, CA, USA). Thapsigargin (Tg) (Millipore Sigma, Burlington, MA, USA) was used to inhibit the SERCA pumps, calpeptin (Selleckchem, Houston, TX, USA) was used to inhibit calpain I/II, SKF96365 (Selleckchem, Houston, TX, USA) or ML204 (Selleckchem, Houston, TX, USA) were used to inhibit TRPC channels, and GSK7975A (Millipore Sigma Burlington, MA, USA) was used to inhibit CRAC channels.

Vero-E6 Cell Culture for Compound Screening

NLRP3 Inflammasome Inhibition in Mice

Male C57BL/6J mice (2 months old, weighing 20–25 g) were purchased from the Experimental Animal Center of Anhui Medical University [SCXK (Wan) 2016-0009]. The mice were kept in a room with constant temperature (22 ± 2°C), appropriate relative humidity (45–65%), and well-ventilated conditions. A 12 h light/dark cycle was maintained, and the mice were allowed free access to food and water. The mice were adaptively fed for one week before the experiments. The experimental procedures were approved by the Ethics Committee of Anhui University of Chinese Medicine, Hefei, China (Approval No. AHUCM-mouse-20210205).
Calpeptin was purchased from Selleckchem (S7396, HPLC > 97%, USA). N-Acetyl-L-cysteine (NAC) was purchased from Beyotime Biotechnology (S0077, HPLC > 97%, Shanghai, China). MCC950 sodium, a selective NLRP3 inflammasome inhibitor, was purchased from Selleckchem (S7809, HPLC > 99.57%).
