Bamboo Leaf Antioxidant Evaluation

One-year-old bamboo (Phyllostachys pubescens Mazel) leaves were collected from a mountain in Xihu District, Hangzhou, Zhejiang Province of China. All the raw materials were smashed into about 200 μm (all the particle sizes were measured on a Topsizer Plus laser particle size analyzer, Zhuhai OMEC Instruments Co., Ltd., Zhuhai, China) after oven drying. Commercial assay kits for the determination of SOD, catalase (CAT), GSH-Px, and methylenedioxyamphetamine, (MDA) including Cell Counting Kit-8 (CCK-8), and BCA Protein Assay Kit, Radio Immunoprecipitation Assay (RIPA) lysis buffer, and Minimum Eagle’s medium (MEM) were purchased from Beyotime Biotechnology Co. Ltd., Shanghai, China. Reactive Oxygen Species Assay Kit (DCFH-DA) was from MedChemExpress (Shanghai).

Facile Synthesis of Functional Materials

All chemicals, including glucose, THF, pyrazine, phosphotungstic acid, 1,3‐ propanesultone, and LGA, were purchased from Sinopharm Chemical Reagent Co. and used without further purification. Elemental analyses were performed by using a CHN elemental analyzer (Vario EL cube) and an inductively coupled plasma spectrometer (TCP, Jarrell‐Ash 1100). The 1H NMR spectra were obtained at 400 MHz by using the Hahn echo at RT. The spin rate was 10 kHz. Scanning electron microscopy (SEM; Hitachi S‐4800, accelerating voltage: 5 kV) accompanied by energy‐dispersive X‐ray spectrometry (EDS; accelerating voltage: 20 kV) was used to study the morphology and the elemental distribution. FTIR spectra (KBr discs) were recorded by using a Nicolet 360 FT‐IR instrument in the 4000–400 cm−1 region.
The acidity of the solid samples was measured by using a Hammett acidity analysis according to a literature procedure.22 The Hammett indicator (p‐nitroaniline, 25 mg) and the catalyst (50 mg) were added to deionized water (10 mL and the mixture was stirred at RT for 12 h, then the solid was separated by centrifugal filtration. The solution was measured by using a UV spectrophotometer at λ=380 nm. Each test was done in triplicate and the reported data is the average value.

Preparation of HER2-targeted PLGA Nanoparticles

C3F8 gas was purchased from Shanghai Renjie Ling Optics Instrument Co., Ltd (Shanghai, China).

Cholesterol Esterase Activity Assay

EV Isolation and Characterization Protocol

EVtrapTM beads were provided by Tymora Analytical Operations (West Lafayette, IN). The automated magnetic bead separation and corresponding “96-well plate” of the EVrich system was in-house built. Lithiumdodecyl sulfate (LDS) loading buffer, tritonX-100 (Shanghai (China) Biotechnology Engineering Co., Ltd.), phosphate-buffered saline (PBS, Gibco), 15% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE, Shanghai (China) EpiZyme Biotechnology Co., Ltd.), anti-CD9 (13403S, CST), anti-CD81 (ab79559, Abcam), anti-TSG101 (ab125011, Abcam), HSP70 (66183, Proteintech), Calnexin (ab133615, Abcam) and polyvinylidene fluoride (PVDF) membranes (Millipore, Sigma) were used in EV capture and western blotting experiment. Sodium deoxycholate, sodium lauroylsarcosinate, Tris (2-carboxyethyl) phosphine, 2-Chloroacetamide, and phosphatase inhibitor cocktail were all purchased from Millipore-Sigma to prepare EV lysis solution. Pierce™ BCA Protein Assay Kit was purchased from Thermo Scientific (23250, reducing agent compatible).
