Enzymatic Activity Assay for Left Ventricular Tissue

Left ventricular tissue was homogenized in PBS on ice and centrifuged at 4°C, at 10,000 rpm for 10 min. The supernatant was collected for measurements. Quantification of ALDH2 and Catalase Activity

Evaluation of Catalase and ALDH2 Activity

Alda-1 and Necroptosis Inhibition Protocol

Primers for ALDH2, RIP1, RIP3, MLKL and GAPDH were acquired from Sangon Biotech Co., Ltd. (Shanghai, China; sequences are presented in Table I). A RevertAid RT Reverse Transcription kit was purchased from Thermo Fisher Scientific, Inc., (Waltham, MA, USA). The SYBR® Premix DimerEraser™ (Perfect Real Time) was acquired from Takara Bio, Inc. (Otsu, Japan).

ALDH2 Activity Assay in Rat Aorta

ALDH2 activity was determined in mitochondria isolated from rat thoracic aorta following solubilization and extraction as specified in the manufacturer's recommendations (mitochondrial ALDH2 activity assay kit; Abcam, Cambridge, UK). The homogenate was then incubated on ice for 20 min and centrifuged at 16 000× g for 20 min at 4°C. Protein concentration of the supernatant was determined and 20 μg of protein was used to detect ALDH2 activity. In this assay, the generation of NADH is coupled to the 1:1 reduction of a reporter dye to yield reaction product concentration, which was monitored by measuring the absorbance increase at 450 nm.
