Tat-linked PKC Inhibitor Peptides in Cardiomyocytes

Tat-linked PKC inhibitor peptides were used at a concentration of 100 nM as described previously (14 (link), 25 (link), 31 (link)). Cardiomyocytes were preincubated with PKCα and β-specific Tat-linked inhibitor peptides for 15 min prior to the protocol outlined in each figure. PKCα and β were also pharmacologically inhibited in isolated cardiomyocytes using Gö6976 (300 nM: Tocris, Bristol, United Kingdom) and LY379196 (300 nM; a kind gift from Eli Lilly, IND, United States) which were perfused with the NT solutions as indicated in the figures. Equivalent concentrations of vehicle (DMSO) had no effect on cardiomyocyte function.
