Caspase-1 Activity Assay in Adipose Cells

The caspase-1 activity in the lysates of SVF, 3T3-L1 preadipocytes and adipose tissue preadipocytes was determined using a caspase-1 fluorometric kit (Biovision, San Francisco, CA, USA) as previously described [44 (link)]. Cells were lysed in hypotonic cell lysis buffer on ice for 10 min and centrifuged to remove the insoluble fraction (12,000× g, 10 min). A total of 50 μL SVF lysate was incubated with 50 μM peptide YVAD-AFC and reaction buffer (50 μL) for 2 h. The fluorescence of the cleaved substrate was measured using a fluorometer at set time intervals.
