Ni-NTA purified L27 Δ 2–9 His6 protein samples were loaded onto a 16.5% Tris-tricine precast gel (Biorad). The protein was electroblotted to a 0.45 μm PVDF membrane for 4 hours at 300 mA in a Criterion Tank Blotter (Biorad). The membrane was stained with Coomassie and the appropriate band was marked, coded and sent to the Iowa State University Protein Facility. There the protein samples on the membrane were washed six times with deionized water and loaded onto a 494 Procise Protein Sequencer/140C Analyzer (Applied Biosystems, Inc).

Ni-NTA Purified Protein Sequencing

Check if the same lab product or an alternative is used in the 5 most similar protocols
Ni-NTA purified L27 Myc-His6 protein samples were loaded onto a 16.5% Tris-tricine precast gel (Biorad) and run until the 10kDa band of the Precision Plus Protein Prestained Standards (Biorad) approached the bottom of the gel. These proteins were electroblotted to a 0.45 μm PVDF membrane for 4 hours at 300mA in a Criterion Tank Blotter (Biorad). The membrane was then stained with Coomassie and the appropriate bands were marked and coded and sent to the Iowa State University Protein Facility for Edman degradation. There the protein samples on the membrane were washed six times with deionized water and loaded onto a 494 Procise Protein Sequencer/140C Analyzer (Applied Biosystems, Inc).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
