Nimesulide

The 1040 compounds used for the small molecule screen were from the Prestwick Chemical Library (Illkirch, France). The following compounds were obtained from Cayman Chemical (Ann Arbor, MI): TWS119 (#10011251), ipsapirone (#22075), bromocriptine (#14598), urapidil (#29004), trimipramine (#15921), dichlorphenamide (#23658), nimesulide (#70640), and mesalamine (#70265). Lithium chloride (LiCl) was obtained from Sigma (St. Louis, MO, #62476), lithium acetate was obtained from Aldrich Chemical Company (Milwaukee WI, #21,319–5), and bortezomib (BTZ) was obtained from EMD Millipore (#179324-69-7).

Celecoxib, NS-398, SC-58125, SC-236, nimesulide, and SC-560 were purchased from Cayman Chemical (Ann Arbor, MI, USA). GCDCA, dimethyl sulfoxide (DMSO), L-NAME, MDL-12330A, and indomethacin were from Sigma-Aldrich (St. Louis, MO, USA). COX-1 siRNA, COX-2 siRNA, and scrambled siRNA were purchased from Santa Cruz Biotechnology, Inc. (Santa Cruz, CA, USA). Monoclonal antibody (MAb) against mouse COX-1 and polyclonal antibody against rabbit COX-2 were obtained from Abcam (Cambridge, MA, USA). Mouse MAb against HA tag was purchased from OriGene (Rockville, MD, USA). Synthetic PGE₂ was purchased from Tocris Bioscience (Ellisville, MO, USA). The anti-PSaV capsid MAb and the anti-PSaV VPg polyclonal antibody were previously described (41 (link)). The secondary antibodies used were horseradish peroxidase-conjugated goat immunoglobulin against rabbit IgG (Cell Signaling, Beverly, MA, USA) and mouse IgG (Santa Cruz) and fluorescein isothiocyanate (FITC)-conjugated goat immunoglobulin against rabbit IgG (Jackson Immuno Research Laboratory, West Grove, PA, USA).

(+) Methamphetamine (Meth) (Sigma, Cat. M-8750) was dissolved in 0.9% sterile saline containing 0.001% heparin (70 U/ml). The dose of Meth infusion was based on previous studies of self-administration and investigator-administered Meth^{6 (link),13 ,52 (link)}.
Nimesulide (7.5 mg/kg, ip, Cayman chemical, Cat. 70640) was injected twice daily as a suspension in 0.9% saline, once in the morning and once in the evening starting on the first day of abstinence when the rats were removed from the self-administration chamber. The dose and time-course of treatment was based on elimination half-life, toxicity and previous studies^{41 (link),53 (link)–57 (link)}.
