Cdc42 antibody

Manufactured by Cell Signaling Technology

The Cdc42 antibody is a research tool used to detect and study the Cdc42 protein in cells. Cdc42 is a member of the Rho family of small GTPases, which are involved in regulating various cellular processes, such as cell shape, motility, and division. The Cdc42 antibody can be used in techniques like Western blotting, immunoprecipitation, and immunocytochemistry to identify and analyze the Cdc42 protein in biological samples.

Immunoblotting Analysis of Cell Signaling

Check if the same lab product or an alternative is used in the 5 most similar protocols
The protein lysates (30–50 μg/sample assessed by BCA protein assay, Pierce Chemical Co., IL) were subjected to 10% SDS-Tris glycine gel electrophoresis and transferred to polyvinylidene difluoride (PVDF) membranes (Millipore, USA). The membranes were incubated with polyclonal GIT1 antibody (Cell Signaling, 1:1000), Rac1 antibody (Millipore 1:1000), Cdc42 antibody (Cell Signaling, 1:1000), RhoA antibody (Cell Signaling, 1:1000), MYO18A antibody (Santa Cruz. 1:1000), SNX27 antibody (Santa Cruz. 1:1000) and α-tubulin (Cell Signaling, 1:5000), at 4°c overnight. Peroxidase conjugated anti-rabbit antibody (1:5000) was used as a secondary antibody. The membranes were developed by Renaissance protein detection kit (DuPont NEN, MA, USA).

Quantifying Activated Cdc42 in HUVECs

Check if the same lab product or an alternative is used in the 5 most similar protocols
HUVECs were seeded at a density of 2 × 105 in a 100 mm dish. The next day, cells were transfected with empty vector, DIAPH1 WT, or DIAPH1 double negative mutant for 24 hr and further starved overnight. Then, cells were treated with PBS or CCN1 (10 ng/mL) for 30 min. Protein lysate was collected by centrifugation at 16,000 × g at 4C for 15 min. Active Cdc42 was extracted from the lysate per the manufacturer’s protocol. Briefly, an equal amount of protein was used for GTPase assay (Active Cdc42 detection kit, 8819, Cell Signalling Technology). GST-PAK1-PBD fusion protein was used to bind the activated form of GTP-bound Cdc42. The level of activated Cdc42 was checked by WB with Cdc42 antibody (Cell Signalling Technology).

Detecting Active Cdc42 in Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Western blot analyses for specific protein targets were performed on freshly prepared tissue or cell lysates following procedures previously described (19 (link)). To detect active Cdc42, GTP-bound Cdc42 was pulled down using GST-human PAK1-PBD supplied by Cdc42 activation kit (Cell Signaling, Catalog # 8819), and detected by Cdc42 antibody (Cell Signaling, #2466). Experiments were repeated on independent biological samples for multiple times.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
