Quantifying Rat Cytokine/Chemokine Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
The collected ACM was used to quantify TNF, IL-6, IL-1β, and IFNγ using the MILLIPLEX MAP RAT cytokine/chemokine magnetic bead panel RECYTMAG-65K (Merck), following the manufacturer’s instructions. Data acquisition and analysis were performed in the LUMINEX platform using the Q-View (Quansys Biosciences) Software.

Plasma Cytokine and Chemokine Profiles

Check if the same lab product or an alternative is used in the 5 most similar protocols
Enzyme-linked immunosorbent assay (ELISA) kits (InvivoGen) were used to measure IFN-α and IFN-β in the plasma following the manufacturer’s instructions. The plates were scanned with FLUOstar Omega microplate reader. Limits of detection: IFN-α, 7.8–500 pg/ml and IFN-β, 15.6–1000 pg/ml. A Q-plex array (Quansys Biosciences) was used to determine cytokine and chemokine levels in plasma. The plate was scanned with Odyssey Infrared Imaging System (LI-COR Biosciences) and analyzed with Q-View (Quansys Biosciences). Limits of detection: TNF, 4.12–3000 pg/ml; IL-1β, 19.2–14000 pg/ml; IL-5, 6.86–5000 pg/ml; IL-6, 6.86–5000 pg/ml; IFN-γ, 10.97–8000 pg/ml; CCL1, 5.6–4000 pg/ml; CCL2, 4.12–3000 pg/ml; CCL22, 5.6–4000 pg/ml.

Multiplex Protein Quantification Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Quantitative protein analysis was performed using a multiplex, antibody-based assay (Q-Plex Human Angiogenesis; Quansys Biosciences, Logan, Utah) in accordance with the manufacturer’s recommendations. A high-resolution image was acquired (Q-View Imager; Quansys Biosciences) and the protein concentration within each PL sample was determined by comparing chemiluminescent intensities with that of an eight-point standard curve (Q-View; Quansys Biosciences).

Cytokine and Chemokine Profiling in SUDV Response

Check if the same lab product or an alternative is used in the 5 most similar protocols
The levels of secreted cytokines and chemokines were analyzed as previously described [13 ]. Cytokine and chemokines IL-2, IL-4, IL-5, IP-10, IFN-γ, and TNF-α were measured in the culture medium of PBMCs following SUDV and SUDV proteins GP, NP, VP30 and VP40 stimulation using Q-Plex technology (Quansys Biosciences, Logan, Utah, USA) according to the manufacturer's instructions. Data were collected with a Quansys Imager (Quansys Biosciences) and results analyzed using Q-View (Quansys Biosciences).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
