Quantitative Growth Factor Analysis

VEGF, IGF-1, bFGF and SDF-1 were quantitatively evaluated by ELISA using the Quantikine system (R&D Systems Inc., Minneapolis, Minnesota). According to the manufacturer’s protocol, standards and pretreated samples were pipetted into the 96 wells. After washing away any unbound substances, an enzyme-linked polyclonal antibody specific for each growth factor was added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution was added to the wells and colour developed in proportion to the amount of growth factor bound in the initial step. The colour development was stopped and the intensity of the colour was measured using a microplate reader set to 450 nm. The obtained values were used to calculate the target protein concentration from a standard curve.

Quantifying Inflammatory Cytokines in Mice
