Fucoidan Extraction and Characterization

Fucoidan standard extracted from U. pinnatifida (Fstd) was purchased from Sigma-Aldrich (Buchs, Switzerland). Sugar standards including L-fucose, D-galactose, and D-glucose were purchased from Macklin (Shanghai, China). Synthetic antioxidants BHA, sodium hydroxide (≥99.0%), and ascorbic acid were also purchased from Macklin (Shanghai, China). Anhydrous ethanol (≥99.0%), hydrochloric acid (4 M), trifluoroacetic acid (≥99.0%), ferrous sulfate (9 mM), hydrogen peroxide (0.3%), ethanolic solution of salicylic acid (5 mM), potassium ferricyanide, 1,1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-hydrazyl (DPPH) and dialysis bags were purchased from Solarbio (Beijing, China). All other chemicals used in this study were of analytical grade.

Histopathological Analysis of Renal Cell Carcinoma

The tissue specimens were fixed in 10% neutral formaldehyde for 16-18 h at room temperature, dehydrated through graded ethanol (50, 70, 80, 90 and 100%), transparentized in xylene at 35°C for 30 min and embedded in paraffin at 60°C for 30 min. Subsequently, the specimens were sliced into serial sections at a thickness of 5 µm and allowed to stand at 45°C. Next, the sections were extracted, baked at 60°C for 1 h, deparaffinized by washing with xylene and rehydrated. Then, H&E staining was performed at a temperature of 35°C as follows: the sections were stained by hematoxylin for 10 min, washed with water for 15 min, differentiated with 1% hydrochloric acid-ethanol for 3 sec, washed with water for 10 sec and stained with 0.5% eosin for 3 min (Beijing Solarbio Science & Technology Co., Ltd., Beijing, China). Then, the sections were dehydrated with graded ethanol, transparentized with xylene and sealed with neutral gum. The pathological changes of the RCC tissues and adjacent normal tissues were observed under an optical microscope (magnification, ×200) (CX31-LV320; Olympus Corporation, Tokyo, Japan).

Bioactive Compound Extraction from Hemerocallis citrina

ABTS and FRAP assay kits were purchased from Suzhou Keming Biotechnology (Suzhou, China). Distilled water was used in these experiments.

Biomimetic Hydroxyapatite-Based Composites

Isolating Trichinella spiralis Larvae

The T. spiralis isolate ISS534 was maintained by serial passaging in ICR mice in the laboratory of animal care in the center of Jilin Agricultural University (Jilin, China). Individual T. spiralis larvae were isolated from mice infected for more than 35 dpi by digesting mouse carcasses with 1% HCl and 1% pepsin (1:3,000; Solarbio, Beijing, China; Li et al., 2010 (link)). Experimental mice were orally administered 350 muscle larvae. The 8-week-old male BALB/c mice were obtained from Hua Fu Kang Biotechnology (Beijing, China) and allowed to eat and drink freely until the end of the experiment. The use of mice in this study was approved by the Animal Ethics Committee and Institutional Life Science Ethics Committee of Jilin Agriculture University (permit no. JLAU 20200904001). The experiments were carried out in strict accordance with the National Laboratory Animal Welfare Guidelines. If any animal has labored breathing, bleeding diarrhea, or becomes moribund it will be euthanized immediately by CO2 inhalation.

Purification of Periplocoside E from P. sepium

EAH sepharose 4B was purchased from general electronics (GE) Healthcare (Beijing, China).
