Quantifying Galectin-1 and Cytokines

Check if the same lab product or an alternative is used in the 5 most similar protocols
Galectin-1 was measured using the Human Galectin-1 Quantikine ELISA Kit (R&D Systems, Minneapolis, MN, USA) according to the manufacturers' instructions. Intra-assay and inter-assay coefficient of variation were 7.1% and 9.5%, respectively. Cytokines were measured in serum using the ultrasensitive multiplex electrochemiluminescence immunoassay (ELISA) (Meso Scale Diagnostics (MSD), Rockville, MD, USA). The human V-PLEX Pro-Inflammatory Panel 1 Human Kit (#K15049) was used to quantify interferon gamma (IFN-γ), interleukin (IL)-1β, IL-2, IL-4, IL-6, IL-8, IL-10, IL-12p70, IL-13 and tumor necrosis factor-alpha (TNF-α) according to the manufacturer's instructions.
Routine analyses of glucose, insulin, and cholesterol were performed at the Department of Clinical Chemistry, Sahlgrenska University Hospital, on a Cobas instrument (Roche Molecular Diagnostics). The homeostasis model assessment for insulin resistance (HOMA-IR) was calculated using the formula [glucose]*[insulin]/22.5 [7 (link)].

Cytokine Profiling in Mouse Serum and Cell Culture

Check if the same lab product or an alternative is used in the 5 most similar protocols
Protein levels of IL-1β, IL-6, and TNF-α in mouse serum were detected using the corresponding mouse ELISA kit according to the manufacturer’s instructions (BioLegend, San Diego, CA, USA; http://www.biolegend.com/). The protein level of Galectin-1 in the cell culture supernatants was detected using Human Galectin-1 Quantikine ELISA kit (R&D Systems, Catalog # DGAL10; https://www.bio-techne.com).

Galectin-1 Serum Levels in CLL Patients

Check if the same lab product or an alternative is used in the 5 most similar protocols
In the experiment, sera of healthy donors and CLL patients with clinically manifested immunodeficiency were used. Previously, Gal-1 serum level was determined using the Human Galectin-1 Quantikine ELISA kit (R&D Systems, Minneapolis, Minnesota, USA) according to the instructions provided. The demographic characteristics of the study participants are given in Table 1.

Soluble Galectin-1 Levels in Metastatic Melanoma

Check if the same lab product or an alternative is used in the 5 most similar protocols
Before patient enrollment, the Bioethical Committee issued a positive decision about this pilot study, and the signed informed consent from all patients was obtained. Nine enrolled patients had been diagnosed with metastatic melanoma with BRAF mutation. All patients received a combination of BRAF + MEK inhibitors, no grade 3–5 toxicity acc. CTCEA were observed during treatment. The patients’ sera were collected before the first administration of BRAF + MEK inhibitors and every 1–6 months from initiation of treatment at the time of radiological and clinical assessment. Radiological assessment was done according to RECIST 1.1. Patients’ characteristics and therapy response status are presented in Table S3. Soluble Gal‐1 in patients’ plasma was quantified using a commercial ELISA kit (Human Galectin‐1 Quantikine ELISA Kit; R&D systems, Minneapolis, MN, USA), according to the manufacturer’s protocol. Serum was diluted ten times, and all samples were analyzed in duplicates.

Quantification of Galectin-1 and PGE2

Check if the same lab product or an alternative is used in the 5 most similar protocols
The gal-1 level in serum (10-fold dilution) or culture media (2-fold dilution) were determined by Human Galectin-1 Quantikine ELISA Kit (R&D Systems), PGE2 concentration in cell culture media was determined by Prostaglandin E2 ELISA Kit (abcam) according to the protocol recommended by manufacturers.

Quantification of Galectin-1 in BALF

Check if the same lab product or an alternative is used in the 5 most similar protocols
Proteins in the BALFs were separated on SDS-PAGE gels (4– 15%; Bio-Rad Laboratories, Hercules, CA, USA) and transferred to PVDF membranes (Millipore, Bedford, MA, USA). Membranes were blocked with 5% milk in TBS and blotted with anti-Gal1 antibody (Santa Cruz Biotechnology, Santa Cruz, CA, USA), followed by HRP-conjugated anti-mouse IgG (Pierce, Rockford, IL, USA). Blots were developed with SuperSignal West Pico chemiluminescent substrate (Pierce). The concentration of galectin-1 in BALFs was also determined with Human Galectin-1 Quantikine ELISA Kit (R&D Systems) according to the manufacturer’s recommendation.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
