Immunocytochemistry of Pancreatic Islet Cells

Functional islets in all groups were detected using insulin immunocytochemistry. IsC or IPCt islets were cultured on glass coverslips (Electron Microscopy Sciences 72290-02 and 72291-01) for immunofluorescence staining. Cells were fixed in 4% paraformaldehyde for 20 minutes, washed with PBS containing 0.3% Triton X-100 followed by incubation with a rabbit anti-insulin antibody (1:1000; Immunostar, USA), mouse anti-α-SMA antibody (1:500; Novus Biologicals, USA), rabbit anti-GFAP antibody (1:500; Proteintech, USA), rabbit anti-glucagon antibody (1:400; Cell Signaling Technology, USA) or rabbit anti-Pdx antibody (1:500; Cell Signaling Technology, USA).
