MTT Cell Viability Assay in HT29 Cells

HT29 cells were cultured overnight in 96-well plates (3.0×103 cells per well) and then treated with various concentrations of compounds in the normal or glucose-free medium for 48 hours. Then the medium was replaced with fresh normal medium, and the cells were cultured for a further 16 hours. Subsequently, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) (Sigma-Aldrich) solution (0.5 mg/mL) was added to the culture medium. After a 4-hour incubation at 37°C, the medium was removed and DMSO was added (100 μL/well); then, absorbance of each well was determined at 570 nm by Multiskan JX plate reader (Thermo Scientific, Waltham MA, USA). Relative cell survival (mean ± standard deviation [SD] of triplicate determinations) was calculated by setting each of the control absorbance from nondrug-treated cells at 100%.
