Lipid Formulation Reagent Sourcing

Efinaconazole Topical Formulation Development

Butylated hydroxytoluene (BHT), disodium ethylenediaminetetraacetic acid (EDTA), isopropyl myristate, isopropyl palmitate, Labrafil®, Labrasol®, N-Methyl-2-pyrrolidone, polyethylene glycol 400 (PEG 400), polyvinylpyrrolidone K30 (PVP K30), propylene glycol, Transcutol® P, and Tween 80, were obtained commercially (Sigma Aldrich, St. Louis, MO, USA). Ethyl alcohol, isopropyl alcohol, acetonitrile, and potassium dihydrogen phosphate were obtained from Qualikems, New Delhi, India.

Solubility Screening of Flavonoids

To select the appropriate surfactant and cosurfactant, the solubility of flavonoids was measured in Cremophor EL (polyoxyethylene [35] castor oil) and TWEEN® 80 (polyoxyethylene [20] sorbitan monooleate; Sigma-Aldrich Co., St Louis, MO, USA), surfactants, and in Transcutol® P (diethylene glycol monoethyl ether) and polyethylene glycol (PEG) 400 cosurfactants. Excess TFRA was added to 1 mL of each solvent at a temperature of 37°C±0.5°C, stirred with a magnetic bar at 300 rpm for 24 hours, and then centrifuged (MiniSpin®; Eppendorf, Hamburg, Germany) at 10,000 rpm for 10 minutes. The supernatant was diluted and assayed using HPLC.

Withania coagulans Fruit-based Formulation

Withania coagulans fruit was purchased from the local market. Pregabalin and Carbopol 934 were generously gifted by Brookes Pharma (Pvt.) Ltd., Karachi and Saffron Pharmaceuticals (Pvt.) Transcutol® P and Tween 80 were acquired from Gattefosse, France (Nanterre, France) and Sigma-Aldrich (St. Louis, MO, USA), respectively. Xylazine and Ketamine were used to anaesthetize the animals. All other chemicals and reagents used were of analytical grade. Distilled water was used during the whole study.

Bovine Lactoferrin-Fenofibrate Nanoemulsion

was procured from lactoferrin.Co, Australia. Fenofibrate IP (white

provided by Sun Pharma Laboratories Limited, India. SR Crodamol GTCC
(super-refined synthetic medium chain triglyceride, colorless, odorless
liquid, specific gravity: 0.95, viscosity: 25 mPass) was received
as a generous gift from Croda India Ltd., Mumbai. Tween 80 was procured
from Research-lab Fine Chem Industries, Mumbai. Edible grade sunflower
oil, soyabean oil, and castor oil are procured from local market.
Transcutol P was procured from Sigma-Aldrich, USA. All other reagents
used in this research were of chemical and analytical grade. Double
distilled water was collected from the distillation system of the
research lab.

Phosphate-buffered saline (PBS), fetal bovine serum (FBS), antibiotics (penicillin/streptomycin), and Dulbecco’s modified Eagle’s medium (DMEM) were purchased from HyClone (Logan, UT, USA).
