Homogeneous Matrix Layer Formation

A thin matrix layer was applied to the surface using an airbrush with a 0.2-mm nozzle (Procon Boy FWA Platinum, Mr. Hobby, Tokyo, Japan) [19 (link)]. The matrix was sprayed repeatedly for 15 sec at 15-sec intervals (for approximately 15 minutes) until 500 μL of 2,5-dihydroxybenzoic acid (2,5-DHB; Bruker Daltonics) solution (50 mg/mL in 70% methanol/0.1% trifluoroacetic acid) was used. During spraying, the distance between the nozzle and the tissue surface was maintained at 15 cm. Pulsed spraying resulted in the formation of a homogeneous matrix layer on the tissue surface.

MALDI-TOF Analysis of Compound Solutions

MALDI-TOF
measurements: First, 1 μL of a DMSO solution of the compound
was spotted on a 384-MTP target plate (Bruker Daltonics, Bremen, Germany)
and air-dried. Subsequently, 1 μL of 2,5-dihydroxybenzoic acid
(2,5-DHB; Bruker Daltonics) matrix (20 mg/mL in ACN/water; 50:50 (v/v))
was applied on the plate, and the spots were left to dry prior to
MALDI-TOF analysis. An UltrafleXtreme MALDI-TOF (Bruker Daltonics),
equipped with Smartbeam-II laser, was used to measure the samples
in the reflectron positive ion mode. The MALDI-TOF was calibrated
using a peptide calibration standard prior to measurement.

Calibrant mixture (Protein Calibration Standard I and Peptide Calibration Standard II), a-cyano-4-hydroxycinnamic acid (a-cyano), and 2,5-DHB were purchased from Bruker Daltonics. The SA was purchased from Sigma. All other reagents were of spectrometry and chromatography grade.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
