Investigating HAUSP Ubiquitination Activity

Flag-HAUSP wild type and K443R mutants were overexpressed respectively in 293T cells and immunoprecipitated by anti-Flag agarose (Sigma). The v5-HectH9 was overexpressed in 293T cells, immunoprecipitated by anti-v5-antibodies (Invitroen) and eluted by v5 tag peptide (10 μg ml⁻¹) in the PBS. The commercial ubiquitination kit (Enzo Life Science, BML-UW9920) was used to perform this experiment. The reaction was incubated for 4-8 h at 37 °C and subjected to Western blot analysis by using anti-K63 ubiquitin antibodies.

In Vitro Ubiquitination of TGF-β1

The in vitro ubiquitination assay was performed as described previously^{50 (link)}. Briefly, Flag-TGF-β1WT, Flag-TGF-β1K315R, or Flag-TGF-β1T282A was transiently overexpressed with His-CHIP in HEK293T cells and purified using anti-Flag magnetic beads (M8823, Sigma–Aldrich) or anti-His magnetic beads (D291–11, Medical & Biological Laboratories, Tokyo, Japan) according to the manufacturer’s protocol.
