CFSE-based Canine Lymphocyte Proliferation

Lymphocyte-enriched mononuclear cells were isolated by gradient separation from heparinized blood in Nycoprep 1.077A (Axis-Shield PoC AS, Dundee, UK) tubes centrifuged at 800g for 30 min at 20°C. After washing twice in PBS with 0.1% human albumin (Grifols Therapeutics, Inc., Research Triangle Park, NC, USA), cells were incubated with 1 μM fluorescent cell staining dye carboxyfluorescein diacetate succinimidyl ester (CFSE) using the CellTrace CFSE Cell Proliferation Kit (Invitrogen, Carlsbad, CA, USA). Duplicates of 4x105 cells were added to each well of a round bottom 96-well plate containing either 0, 1, or 100 pg/mL of staphylococcal enterotoxin A (SEA; Sigma, St. Louis, MO, USA) and cultured for 7 days. Cells were harvested and labeled with an AlexaFluor 647-conjugated monoclonal anti-canine CD18 antibody CA1.4E9 (AbDSerotec, Raleigh, NC, USA) and analyzed by flow cytometry for CD18 expression (FL4 channel) and CFSE staining (FL1 channel, BD FACSCalibur, Becton-Dickinson, San Jose, CA, USA).

PBMC Stimulation and IL-2 Measurement

Human peripheral blood mononuclear cells (PBMC), isolated from healthy donor leukocones (NHSBT) using Ficoll-Paque Plus (GE Healthcare), were cultured at 2 × 105 cells/well in 96-well plates (Corning) pre-coated with 0.5 μg/mL anti-human CD3 monoclonal antibody (OKT3, eBioscience). Cultures were supplemented with 100 ng/mL staphylococcal enterotoxin A (SEA) (Sigma), 30 μg/mL tremelimumab or isotype control (MedImmune) and selumetinib at the concentrations indicated. Following 72 h incubation at 37 °C, IL-2 release was measured by DELFIA using reagents from a human IL-2 ELISA kit (R&D systems) and europium-labelled streptavidin (Perkin-Elmer). Results were repeated in two independent experiments.

TRAM 34 (#2946) was purchased from R&D Systems and Venetoclax (ABT-199, #ab217298) from Abcam, Rituximab (lot oN7370B16) was from the local pharmacy (Universitätsklinikum Homburg, Hoffmann La Roche). All other chemicals and reagents not specifically mentioned were from Sigma Aldrich, ThermoFisher Scientific.
