Synthesis and Characterization of Doxorubicin-loaded Hydrogel Nanoparticles

Acrylic acid (Sigma-Aldrich, China) was distilled before use. N,N’-methylenebisacrylamide and N-Isopropylacrylamide (J&K Scientific., Ltd., Shanghai, China) were recrystallized from methanol and n-hexane, respectively. Doxorubicin hydrochloride (DOX HCl) was purchased from Xianghe Shunda Fine Chemical Co., Ltd. (Wuhan, China). 1-Ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC.HCl), N-Hydroxysuccinimide (NHS), potassium persulfate, sodium dodecylsulfate, and 5-aminofluorescein were analytic grade and purchased from J&K Technology, Ltd. cRGD was synthesized by ABBiochem Co., Ltd. (Shanghai, China). DMEM and RPMI-1640medium were obtained from HyClone Laboratories, Inc. (Utah, USA). Trypsin was purchased from Gibco-BRL Life Technologies (Carlsbad, CA). Biosharp (Hefei, China) supplied DAPI and MTT. All other chemicals were analytical grade and used without further treatment.

Synthesis of Functional Polymeric Materials

Sodium borohydride and 3-bromo-1-propanol were purchased from Sigma-Aldrich. Acrylic chloride and tellurium powder were purchased from Shanghai Reagent Company. Acrylic chloride was purchased from Anhui Wotu Co., Ltd. and used after being purified by vacuum distillation. N,N′-Methylene diacrylamide (MBA), acrylamide (AM), hexadecyl trimethyl ammonium bromide (CTAB), 5,5′-dithio-bis(2-nitrobenzoic acid), 4-nitrothiophenol (NBT), cumyl hydroperoxide, and hydrogen peroxide were used as received from J&K Scientific Ltd. N-Isopropylacrylamide (NIPAM) was purchased from J&K Scientific Ltd., and used after cyclohexane/methanol recrystallization and drying. 2-Bromopropionyl bromide was purchased from Lancaster. Tris(2-dimethylaminoethyl)amine (ME6TREN) was prepared according to the previously reported methods.51 (link) Triethylamine and tetrahydrofuran were purchased from Nanning Lantian Reagent Co., Ltd., and used after rigorously dried with sodium. 3-Carboxyl-4-nitrobenzenethiol (TNB) was synthesized from 5,5′-dithiobis(2-nitrobenzoic acid) using the previously reported methods.20 (link)

Deionized water, which was purified by a Millipore Milli-Q system, was put to use in all experiments.

Synthesis of NIPAM-based Hydrogels

Silica Wafer Surface Modification

(MPTMS), trichloro(1H,1H,2H,2H-perfluorooctyl) silane (PFS), 2-bromoisobutyryl
bromide, and copper(I) chloride (CuCl) were provided by Aldrich. The
polystyrene microparticles with a diameter of 620 nm were provided
by Wuhan Sphere Scientific Co., Ltd. and were cleaned with water and
50% ethanol several times before use. Dichloromethane, absolute ethanol,
triethylamine, sodium dodecyl sulfate (SDS), methanol, 1,2-dichloroethane,
and the four components of the photoresist were used directly after
received. The masks employed for photolithography were machined by
the Institute of Microelectronics of Chinese Academy of Sciences.
The TJ-1A-Micro Flow syringe pump was purchased from Baoding Longer
Precision Pump Co., Ltd. The water in all experiments was deionized
and doubly distilled prior to use.
