Tensile Stress-Strain Analysis of MXene/CNT Strips

Tensile stress–strain curves of MXene (/CNT) strips (20 mm × 5 mm × t) were recorded at a loading rate of 1 mm min−1 using Instron 5567 tester with a 100 N force cell. The tensile strength was then directly obtained by the maximum stress that the sample film could withstand before breaking. Electron conductivity (σ) was calculated by the thickness (t), and sheet resistance (Rs) was measured with an RTS‐8 four‐probe resistivity meter using σ=1Rs×t. Surface hydrophilicity was tested by a contact angle meter (Biolin Scientific, Attention).

Hydrogel-based Transdermal siRNA Delivery

The PEI-PBA@FAM-siRNA was comixed with PEG polyesters and preheated at 30°C for 10 min to reduce its liquidity. Then, the mixture hydrogel was transferred to a horizontally placed PLGA membrane, while the membrane was placed in a 37°C thermostat for 2 min to prepare lubricated gene-hydrogel patches. The prepared patches were used to evaluate the release of siRNA. First, the patches were placed in the upper chamber of the Transwell cell culture plate, and the release level was assessed by adding an appropriate amount of 1× PBS to the lower chamber. A constant temperature of 37°C was maintained throughout, the PBS in the lower chamber was changed every 24 hours, and the fluorescence intensity in the solution was measured by a fluorescent enzyme marker. The concentration was calculated by measuring the fluorescence intensity of known concentrations of PEI-PBA@FAM-siRNA to draw a standard curve. The contact angles of the prepared patches were determined by a contact angle meter (Biolin Scientific). The patch surface morphology was observed using a SEM (Sirion 200).
