Evaluating Rapamycin's Effect on Oocyte Maturation

For the animal experiment, ICR mice aged 40~48 weeks were purchased from Beijing Vital River Laboratory Animal Technology Co., Ltd. Totally, 18 aged female mice were sacrificed by cervical dislocation and 241 oocytes were obtained. All the experiments were repeated three independent times. Specifically, the bilateral ovaries were isolated, transferred into M2 medium (EasyCheck, Nanjing, China) containing IBMX (Selleck, Houston, TX, USA), and punctured with sterile needles to release immature oocytes. After being washed five times in M2 medium, the denuded GV oocytes were moved to the IVM medium (EasyCheck, Nanjing, China), with or without 10 nM rapamycin (MedChemExpress, Monmouth Junction, NJ, USA), and then cultured for 16 h at 37°C under 5% CO2 in humidified air. Following IVM, MII oocytes were recognized with the extrusion of the first polar body (PB1), and the maturation rates of the control group and rapamycin group were recorded and compared.

Epididymal Sperm Isolation and Analysis

The cauda epididymis from 3-month-old male mice were placed in 1 ml modified HTF media and backflushing was used to obtain spermatozoa.36 (link) Spermatozoa were incubated with DAPI (1 : 1000, Sigma-Aldrich, St. Louis, MO, USA) for 2 min. Samples were placed in Leja counting chambers (Orange Medical, Rotterdam, The Netherlands), and sperm concentration was evaluated by using the IVOS sperm analyzer (Hamilton Thorne Bioscience, St. Louis, MO, USA). Cauda epididymal spermatozoa in the media were treated with or without 50 μM to 1 mM cAMP analogs, and 100 μM IBMX (all purchased from Selleck, Shanghai, China). Motility of knockout spermatozoa after a 0- to 60-min incubation were analyzed by computer-assisted sperm analysis using the IVOS sperm analyzer (Hamilton Thorne Bioscience) and the motility rate (%) was measured.
