Refill fluids contained PG (Thermofisher, Tustin CA, USA) and VG (Thermofisher, Tustin CA, USA). At 80% confluency, BEAS-2B cells were treated for 24 hours with nicotine, PG/VG, PV/VG with nicotine, or JUUL fluids. All treatments were diluted with BEGM culture medium to reach working concentrations. Liquid (−)- nicotine (Sigma-Aldrich, St. Louis, MO, USA) was diluted to reach final concentrations of 0.03 mg/mL or 0.3 mg/mL. PG/VG (30/70 ratio) was diluted to reach a final concentration of 0.5% in volume. Stock solutions of PG/VG with nicotine were made and then diluted with culture medium to 0.5% PG/VG with either 0.03 mg/mL nicotine or 0.3 mg/mL nicotine. JUUL “Virginia Tobacco” was diluted to 0.5%, which contains PG/VG at 0.5% concentration and nicotine at 0.3 mg/mL.

[125I]-Epibatidine (2200 Ci/mmol), [3H]SCH23390 (N-methyl [3H], 84.3 Ci/mmol), [3H]mazindol (4’-[3H], 20.6 Ci/mmol) and [3H]raclopride (methoxy-[3H], 20.6 Ci/mmol) were obtained from Perkin-Elmer NEN, Boston, MA. α-Conotoxin MII (α-CtxMII) and [125I]-α-CtxMII were prepared as described previously (Whiteaker et al. 2000 (link), Cartier et al. 1996 (link)). Unlabeled 5I-epibatidine was a generous gift from Dr. Kenneth Kellar, Georgetown University, Washington, DC. Liquid (−)-nicotine was purchased from Sigma-Aldrich Chemical Company (St. Louis, MO) and was redistilled periodically. All additional chemicals were of reagent grade.

Unlabeled or uniformly 15N- or13C,15N-labeled Aβ(1–40) peptides were bought lyophilized from AlexoTech AB (Umeå, Sweden). The peptide samples were freshly dissolved and prepared before the measurements, according to previously published protocols61 (link). Liquid (-)-nicotine (Sigma-Aldrich) and acetate salts of Cd(II), Cr(III), Pb(II), and Pb(IV) (Sigma-Aldrich) were dissolved or diluted in 20 mM phosphate buffer. Naphthalene, phenanthrene, pyrene, and B[a]P were first dissolved in 50% DMSO and then diluted in 20 mM phosphate buffer. NaOH/HCl was used to adjust the pH of all stock solutions to 7.35.

Care and Utilization Committee at the University of Colorado, Boulder and were conducted in accordance with the European Community Council Directive (2010/63/UE).
All of mice were bred at the Institute for Behavioral Genetics, and were housed at 22°C with lights on from 7 a.m. to 7 p.m. and allowed free access to food and water. The (±)-[ 3 H]-epibatidine ligand (Epi, specific activity: 66 Ci/mmol) was purchased from Perkin Elmer (Waltham, MA, USA); nonradioactive Epi, nicotine and α-bungarotoxin (α-Bgtx) were purchased from Tocris Bioscience (Bristol, UK). Liquid nicotine was purchased from Sigma/Aldrich (St. Louis, MO) and redistilled periodically. All other reagents (PMSF, proteases inhibithors, chemicals) were also from Sigma-Aldrich.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
