Analytical Standards for Pharmacological Assays

Liquid Chromatography-Mass Spectrometry of Zearalenone Metabolites

Liquid chromatography–mass spectrometry (LC-MS)-grade acetonitrile (ACN) and methanol (MeOH) were obtained from Sigma-Aldrich, and water ( H2O , LC-MS grade) was purchased from VWR International GmbH. Ammonium fluoride (LC-MS grade) was obtained from Honeywell Fluka. ZEN, α-ZEL , β-ZEL , α-ZAL , β-ZAL , and C1813-ZEN were obtained from either Romerlabs or Sigma-Aldrich, whereas ZEN-14-GlcA, α-ZEL-GlcA , and β-ZEL-GlcA were synthesized at the Technical University of Vienna and kindly provided by Dr. Mikula (Mikula et al. 2012 (link), 2013 (link)). ZEN-14-Sulf was a kind gift from Prof. Berthiller from the University of Natural Resources and Life Sciences, Vienna (Vendl et al. 2010 (link)). Antipyrine was obtained from Sigma-Aldrich. All reference standards were dissolved in ACN. ZEN and its metabolites were combined manually to a multianalyte calibration solution (each analyte at a concentration of 10mg/L ). All single and combined analytes were stored at 20°C . ZEN for transfusion experiments was obtained from Toronto Research Chemicals (purity 98.9%).

Permeability Evaluation of Five Drugs

Human iPS-IECMs, which were cultured on the cell culture inserts, were preincubated with HBSS adjusted to pH 7.4 with 10-mM HEPES for 10 minutes at 37°C. The 1.5 mL of HBSS containing 1-μM Antipyrine, metoprolol, fexofenadine, atenolol, and pravastatin was added to the apical chamber, and 2.6 mL of HBSS containing 0.1% DMSO was also added to the basolateral chamber. Antipyrine, metoprolol, fexofenadine, atenolol, and pravastatin were purchased from Sigma-Aldrich. After 30-, 60-, 90-, and 120-minute incubation at 37°C, the solution was collected from the basolateral chamber. Antipyrine, metoprolol, fexofenadine, atenolol, and pravastatin concentration was measured by UPLC-MS/MS. Quantification of Antipyrine by HPLC

Ltd (Shanghai, East China, China. Lot.: 20,221,213). Other chemicals and solvents were of analytical grade and obtained from a commercial supplier.
