Hyperinsulinemic-Euglycemic Clamp in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Survival surgery was performed at 5~6 days before clamp experiments to establish an indwelling catheter in the jugular vein. On the day of the clamp experiment, mice were fasted overnight (~15 hrs), and a 2-hr hyperinsulinemic-euglycemic clamp was conducted in conscious mice with a primed and continuous infusion of human insulin (150 mU/kg body weight priming followed by 2.5 mU/kg/min; Humulin, Eli Lilly & Company, Indianapolis, IN, USA). To maintain euglycemia, 20% glucose was infused at variable rates during clamps. Whole body glucose turnover was assessed with a continuous infusion of [3-3H]glucose (PerkinElmer, Waltham, MA, USA), and 2-7deoxy-D-[1-14C]glucose (2-[14C]DG) was administered as a bolus (10 µCi) at 75 min after the start of clamps to measure insulin-stimulated glucose uptake in individual organs.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
