Plasma was separated from EDTA whole blood by centrifugation at 2000 G for 15 min at 4°C. A 1:50 dilution was prepared using SDS-PAGE loading buffer. Proteins were separated by SDS-PAGE and transferred to nitrocellulose membranes for immunoblotting. Membranes were probed with polyclonal primary Abs: rabbit anti-CSF1 (1:500, MBS551037; MyBioSource) and 1:2000 sheep anti-Transferrin (1:2000, ab9033; Abcam) in PBS/3% milk powder/0.1% (v/v) Tween 20 at 4°C overnight. The secondary Abs used were goat anti-rabbit HRP (1:5000, no. 7074; Cell Signaling Technologies) and rabbit anti-sheep HRP (1:10,000, ab97130; Abcam). Blots were visualized using Pierce ECL Western blotting solution (Thermo Fisher Scientific).

Transferrin Immunofluorescence Staining

Check if the same lab product or an alternative is used in the 5 most similar protocols
293 cells were washed with phosphate-buffered saline (PBS), fixed using 4% paraformaldehyde for 15 min at room temperature, and washed again with PBS. Cells were permeabilized using 0.2% Triton X-100 in PBS for 15 min at room temperature, followed by washing the samples with PBS. Samples were incubated for 1 hour at room temperature in blocking buffer (PBS with 10% bovine serum albumin). Anti-transferrin antibody (Abcam ab9033, England) was used as primary antibody and incubated in blocking for 2 hours at room temperature; after washing with PBS cells were incubated for 45 min at room temperature with an anti-rabbit secondary antibody diluted in blocking buffer [21] .

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
