Adoptive Transfer of JEDI T-cells

CD8+ T-cells were purified from spleens and LNs (cranial, axillar, brachial, inguinal and mesenteric) of JEDI and control littermates by mechanical disruption and filtering through 70μm cell strainer. After RBC lysis, CD8+ T-cells were negatively selected using the mouse CD8+ T-cells isolation kit II from Miltenyi following manufacturer’s instructions. 5×105-5×106 JEDI or control T-cells were transferred per mouse by tail vein injection. For activation of T-cells, recipient mice were injected i.v. with 1×108 transducing units of a vesicular stomatitis virus (VSV)-pseudotyped lentiviral vectors encoding green florescent protein (LV.EGFP).
