Extraction and Fractionation of Hamamelis virginiana

Check if the same lab product or an alternative is used in the 5 most similar protocols
Full extracts (60% ethanol) of Hamamelis virginiana L. leaf or bark, as well as ultrafiltration (UF) concentrates and filtrates were prepared as previously described [47] (link). For bark extract fractionation into UF-concentrate and filtrate, a Pro Flux M12 Tangential Flow Filtration System (Millipore) with Pellicon 2 Ultrafiltration Cassettes (C Screen, Millipore) was used. All extracts and UF-fractions were prepared at Dr. Willmar Schwabe GmbH & Co. KG, Karlsruhe and provided as dry powders. Concentrations indicated designate dry extract weight per volume of solvent. Gallic acid monohydrate (“gallic acid”), penta-O-galloyl-β-D-glucose (“pentagalloylglucose”), hamamelitannin, tannic acid and (−)-epigallocatechin gallate (EGCG) were purchased from Sigma-Aldrich. The buffer capacity of the cell culture medium fully neutralized the acidity of compounds added to in vitro experiments.

Comprehensive Analysis of Rooibos Extracts

Check if the same lab product or an alternative is used in the 5 most similar protocols
Citric acid, disodium hydrogen phosphate dihydrate, and methanol were from VWR International (Leuven, Belgium), acetone and formic acid from Merck (Darmstadt, Germany), gallic acid (≥98% purity), and penta-galloyl-glucose (≥96% purity) from Sigma Aldrich (Steinheim, Germany), luteolin (≥99% purity) and orientin (≥99% purity) from Extrasynthèse (Genay, France), fermented and green rooibos powder extracts, and aspalathin (≥98% purity) from Phytolab (Vestenbergsgreuth, Germany). Analytical grade chemicals from Sigma Aldrich (Steinheim, Germany) and ultrapure water prepared using a Sartorius 611 Ultrapure Water System (Sartorius, Göttingen, Germany) were used throughout the study.

Pentagalloyl Glucose Inhibits TNBC Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
TNBC cells were obtained from American Type Culture Collection (ATCC) (Manassas, VA) and pentagalloyl glucose (purity 96.8%) from Sigma-Aldrich Co. (St. Louis, MO, USA). Human cytokine antibody arrays (Cat# AAH-CYT-6-4), ELISA (Cat# ELH-GRO and Cat# ELH-GRO-alpha), and apoptosis kits (Cat# 68FTAnnV-S100), and tumor necrosis factor-alpha (TNF-α)/human recombinant were obtained from RayBiotech (Norcross, GA, USA). Primers for RT-PCR, SsoAdvanced Universal SYBR (Cat# 1725271), iScript Advanced cDNA Synthesis Kit (Cat# 1725037), apoptosis PCR arrays (Cat# 10034106) were purchased from Bio-Rad (Hercules, CA, USA). Western assays (Cat# SM-W004) were obtained from ProteinSimple (San Jose, CA, USA) and antibodies from Thermo Fisher (Waltham, MA, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
