T Cell Proliferation Assay with MDSC

Naïve WT T cells were purified from spleens as described and labeled with the proliferation tracker Cell Trace Violet according to manufacturer protocol. T cell proliferation was stimulated by adding anti-CD3/CD28 mouse T-activator beads (Thermo) at a 1:1 T cell to bead ratio according to manufacturer protocol. T cells (2×105) were subsequently co-cultured with MDSC at 1:4, 1:8, or 1:16 MDSC to T cell ratios and incubated for 3 days prior to flow cytometric analysis.
