Expression and Purification of Recombinant ADI

The ADI plasmid of Mycoplasma arginini was a kind gift from Dr. Wei-Chiang Shen (School of Pharmacy, University of Southern California, Los Angeles, USA) and was transformed into Escherichia coli (E. coli) BL21. rADI was prepared and purified to homogeneity as previously described [19 (link)]. The reagents for the E. coli culture medium were purchased from Becton Dickinson Transduction Laboratory (Lexington, KY). Ampicillin was purchased from MDBio (Rockville, Maryland). The Micro BCA protein assay reagent kit was obtained from Pierce (Rockford, IL). Isopropyl β-D-1-thiogalactopyranoside (IPTG), dithiothreitol (DTT), 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), sulfanilamide, N-(1-napthyl)-ethylenediamine-2HCl, retinoic acid (RA), 2,3-diaminonaphthalene (DAN), N-methyl-D-aspartic acid (NMDA), (+)-MK-801, and rhodamine 123 (Rho 123) were purchased from Sigma (St. Louis, MO).

Overexpression of Truncated HCV Core Protein

The cDNA sequence of truncated HCV core protein 1–116 (domain 1, p116) was inserted into the pQE30 plasmid, a vector for expressing N-terminally His-tagged recombinant proteins (QIAGEN, Germantown, MD, USA), and transformed into E. coli cells (M15 strain, QIAGEN, Germantown, MD, USA). The bacteria were selected by Luria–Bertani (LB) broth (LAB M, Heywood, England) agar containing 50 μg/mL ampicillin (MDBio Inc., Rockville, MD, USA) and 10 μg/mL kanamycin (Progen, Australia). For core protein overexpression, transformed bacteria were cultured in 2xYT medium containing ampicillin and kanamycin (final concentrations of 50 μg/mL and 10 μg/mL, respectively) at 37 °C. 1 mM isopropyl-D-1-thiogalactopyranoside (IPTG, MDBio Inc., Rockville, MD, USA) was added to the bacteria culture for protein induction when the OD595 of the bacteria culture reached 0.5~0.6. After 3 h of induction, bacterial pellets were collected by centrifugation at 7000 rpm for 20 min.

Cytotoxicity Evaluation of Cisplatin

Cancer cell line culture and maintenance AGS (Purchased from Pricella Biotech.) was cultured in the RPMI 1640 medium (ThermoFisher) and HEK293t/HCT116/ HeLa cells were cultured in the DMEM high-sugar medium (Corning) plus 10% of fetal bovine serum (WISENT) and penicillin-streptomycin (ThermoFisher). HeLa and HCT116 cells were obtained from the National Collection of Authenticated Cell Cultures, and HEK293t and Hep 3B cells were gis from Prof. Fei XIAO (Sun Yat-sen University). All cells were maintained in a humidied cell culture incubator with a 5% CO 2 gas level at 37 °C.

Versatile Nanocarrier Synthesis and Optimization
