Doxorubicin and Drug Preparation Protocols

Hyaluronate-Alginate Hydrogel Synthesis

Adipic Acid Dihydrazide-Modified Hyaluronic Acid Synthesis

ADH-HA was produced by modifying the carboxylic acid moieties of the D-glucuronic acid with adipic acid dihydrazide. Adipic acid dihydrazide (ADH) (≥98%; Sigma-Aldrich, Saint Louis, MO, USA) was added to a 1 wt.% HA (Mw~1.5–1.8 MDa; Sigma-Aldrich, Saint Louis, MO, USA) solution prepared in Milli-Q water, at a molar ratio of 1 (ADH:HA). Then, the reaction occurred in the presence of N-(3-Dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride (≥98.0% Sigma-Aldrich, Saint Louis, MO, USA) (molar ratio 0.25; EDCI:HA) at pH 4.75, by addition of HCl dropwise, at room temperature, until no further pH variation was observed. The solution was then dialyzed firstly against diluted HCl containing 0.1 M NaCl (3 × 5 L, 48 h), followed by dialysis in diluted HCl (pH 3.5) (3 × 5 L, 24 h) and finally, against Milli-Q water. The resultant solution was frozen at −80 °C, recovered by freeze-drying, and stored until further use at −20 °C [11 (link)].

Hydrogel Erosion Kinetics Comparison

ELP-HA (2wt%, 1 wt%) hydrogel was prepared as described in the previous section. A control hydrogel, consisting of 3 wt% HA-ALD, formed at the same stoichiometric ratio of hydrazine to aldehyde groups as the ELP-HA gel, was crosslinked using a small molecule crosslinker, adipic acid dihydrazide (ADH, Sigma-Aldrich). The control gel was formed by mixing the 4.5 wt% HA-ALD stock solution with 4.665 mg/mL ADH in PBS at a 2:1 volume ratio. 30 μL of the desired hydrogels were prepared in separate centrifuge tubes for the erosion test. Four replicates were prepared in each group. The gels were immersed in 1 mL PBS and incubated at 37 °C. 200 μL of the buffer was collected and replaced by fresh PBS at days 1, 2, 3, 6, and 9. Data collection was stopped on day 9 since the control hydrogel was completely degraded. The remaining ELP-HA hydrogels were degraded in 2 mg/mL hyaluronidase to allow determination of remaining hydrogel content for data normalization. Alginate Oligomers and Trimannuronate Synthesis

Guluronic acid oligomers (G oligomer) with two different molecular weight distributions were prepared 10 from partially acid hydrolysed, high guluronate alginate from Laminaria hyperborea stipes by acid precipitation to give oligomers with a number average degree of polymerization (DPn) of 21 and a fraction of guluronic acid (FG) of 0.90 (determined by NMR). Samples with DPn of 4 and 10 were prepared by further hydrolysis.
Trimannuronate was an inhouse sample prepared according to previously described methods. 11, 12 Dextran T-2000 (Mw = 2 000 000 g/mol) was purchased from Pharmacia Fine Chemicals. Maltotriose, trigalacturonic acid, adipic acid dihydrazide (ADH), O,O'-1,3,-propanediylbishydroxylamine dihydrochloride (PDHA) and 2-methylpyridine borane complex (α-picoline borane) was purchased from Sigma-Aldrich. PEG linkers were purchased from BroadPharma, US. All other chemicals were obtained from commercial sources and of analytical grade.

Preparation of PA-OMVs-DT Conjugate

PA-OMVs were covalently conjugated to DT as a carrier protein using the adipic acid dihydrazide (ADH) (Sigma, USA) as a spacer molecule and 1-ethyl-3- (3- dimethyl aminopropyl) carbodiimide (EDAC) (Sigma, USA) as a linker. At first, 2 mg/ml of PA-OMVs was exposed to EDAC (final concentration of 0.1 M), and then reacted with ADH for 5 h at pH: 8. Thin-layer chromatography (TLC) was used to monitor the reaction's solution for 6 h. The conjugation of PA-OMVs and ADH was then purified for 24 h using three water changes and a 2 kDa cut-off dialysis bag against water at 4 °C. Then, 2 mg/ml of this solution reacted with DT containing 1 mg/ml protein in the presence of EDAC for 24 h. Then, PA-OMVs-DT conjugate was purified via Sephadex G-750 gel filtration chromatography (Sigma, USA).
The fractions were collected and centrifuged as a PA-OMVs-DT conjugate after being measured at 280 nm (protein) in the obtained fractions. Finally, the conjugated molecules were lyophilized in glass vials with a volume of 1 ml, passed through a 0.45 mm Millipore filter (USA), and kept at − 20 °C until injection.

Dulbecco's Modified Eagle Medium (DMEM) with GlutaMAX™ and without nucleosides, foetal bovine serum, streptomycin, penicillin, amphotericin B, HEPES Buffer solution, StemPro® Adipogenesis Differentiation kit and StemPro® Chondrogenesis Differentiation kit were obtained from Gibco®. AccutaseTM Cell detachment solution, Stain Buffer and CD90, CD105, CD44, CD34, CD19, CD11b, CD45, MHC Class II antibodies were obtained from BD Biosciences©.
