Upon organ extraction, tissues were fixed in 4% PFA (pH 7.4) for 48 h and subsequently rinsed ×2 in 1× PBS (pH 7.4). Tissues were then stored in 70% ethanol for shipping. Paraffin embedding and assembly of 4.5 mm core tissue microarrays (TMA) was conducted by Pantomics, Inc. (Richmond, CA). Sectioning (5 μm thick) of TMA was conducted at the Histology Department (Chinook Regional Hospital). In brief, upon citrate buffer epitope retrieval, slides were rinsed and subjected to serum blocking to prevent the non-specific binding of immunoglobulins. Sections were then incubated overnight with p38 MAPK rabbit antibody (Cell Signalling #9212). Slides were then washed 3 × 10 min in 1× PBS (pH 7.4), incubated with Alexa Flour 488 goat anti-rabbit antibody (Invitrogen A11034) for 1.5 h and subsequently washed 3 × 10 min in 1× PBS (pH 7.4) before dehydration and counterstaining with 4′,6′-diamidino-2-phenyl-indole, dihydrochloride (DAPI) (Prolong Gold anti-fade reagent with DAPI Invitrogen).
