Zinc-Dependent FluoZin-1 Fluorescence Assay

Reactions were performed in a final volume of 100μl containing 50 μM FluoZin™-1 (ThermoFisher, F24180) and 50 μM ZnCl2 in20 mM Tris pH 7.5, 5% DMSO. THL was added at the indicated concentrations in the presence and absence of 2.5 mM β-mercaptoethanol. FluoZin™-1 fluorescence (Ex/Em: 495/515nm) was measured after 5 mins of incubation in black 96-well plates using an EnSpire multilabel plate reader with a monochromator.
