Duoset human trem 1

Manufactured by R&D Systems
Sourced in United States

The DuoSet-Human TREM-1 is a quantitative sandwich enzyme-linked immunosorbent assay (ELISA) developed for the measurement of human TREM-1 in cell culture supernatants, serum, and plasma. TREM-1 is a cell surface receptor that is upregulated during inflammation and infection.

Metabolic and Inflammatory Biomarkers

We performed blood collection by venipuncture after 12 hours of fasting and without alcohol, coffee, or tea intake for 24 hours. The heparin and plasma samples were separated by centrifugation at 2465 g at 5°C for 15 minutes and stored at -80°C. Serum concentrations (mg/dL) of glucose, total cholesterol, high-density lipoprotein (HDL-c), and triglycerides were analyzed by colorimetric or turbidimetric method. The serum concentration (IU/mL) of insulin was analyzed by electrochemiluminescence. HOMA-IR value was calculated from the fasting insulin and glucose dosages. Plasma concentrations of Ox-LDL and serum levels of the inflammatory molecule sTREM-1 were analyzed by enzyme-linked immunosorbent assay (ELISA), using the specific analysis kit (Mercodia, Uppsala, Sweden) and DuoSet-Human TREM-1 (R&D Systems, Minneapolis, USA).

Quantification of Soluble TREM-1 and Inflammatory Mediators

Check if the same lab product or an alternative is used in the 5 most similar protocols
Soluble (s)TREM-1 was quantified in cell-free Mf supernatants by human TREM-1 DuoSet (R&D Systems). Secreted IL-12, TNFα, IL-1β, CXCL8, IL-6, and IL-10 (Peprotech, Milano), osteopontin (OPN), CCL18, CCL24 and TGFβ1 (R&D Systems, Space Import Export, Milano, Italy) content in Mf CM was also measured by specific ELISA. HMGB1 levels were measured in SF and plasma from OJIA patients and age-matched controls using HMGB1 ELISA kit II (IBL International, Hamburg, Germany); the lower limit of detection was 0.3 ng/mL. Optical density was determined using a Spectrafluor Plus plate reader from TECAN (Milano, Italy). All assays were done in duplicate.

Human TREM-1 DuoSet ELISA Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Human TREM-1 DuoSet (R&D Systems Europe, Abingdon, UK) was performed according to manufacturer’s instruction. When indicated buffer solutions where replaced by buffers (coating and blocking buffer = pcb = protein containing buffer) described above at Radsak ELISA.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
