Nrf2 Activation in Spinal Cord Injury

Check if the same lab product or an alternative is used in the 5 most similar protocols
At 4 and 8 weeks after CSCI, spinal cord tissue was homogenized in IGEPAL CA-630 buffer (Sigma-Aldrich, St. Louis, MO, USA, Cat# 9002-93-1). After centrifugation, the supernatant was subjected to immunoprecipitation with anti-Nrf2 (rabbit, 1:1000, Proteintech, Cat# 16396-1-AP, RRID: AB_2782956) at 4°C overnight. The samples were then incubated with immobilized antibody beads at 4°C for 2 hours. They were also subjected to immunoprecipitation with the IgG isotype control. Following immunoprecipitation and washing with PBS three times, the samples were eluted with glycine-HCl (0.1 M, pH 3.5), and the proteins were subsequently analyzed for Nrf2 and MafG using western blotting.

Protein Extraction from Myocardial Infarction

Check if the same lab product or an alternative is used in the 5 most similar protocols
The infarct area of MI heart specimens was removed on day 7 after MI and homogenized in radioimmunoprecipitation assay (RIPA) buffer (containing 50 mmol·L-1 Tris-HCl, pH 6.8, 150 mmol·L-1 NaCl, 1 mmol·L-1 PMSF, 2 ng·mL–1 aprotinin, 1% Triton X-100, 1% SDS, and 1% sodium deoxycholate). HEK 293 T and H9c2 cells were washed with PBS and lysed with IGEPAL CA-630 buffer containing protease and phosphatase inhibitors (Sigma-Aldrich). The samples were centrifuged and supernatant was transferred to new tubes. Protein concentration was calculated using bicinchoninic acid (BCA) assay (Pierce Biotechnology, Rockford, IL, USA). Unused proteins were stored at −80 °C until further use.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
