Norepinephrine Regulation of Endothelial Cells

A type 1 collagen positive endothelial cell line (BC-1)73 (link) was kindly provided by Dr. M. Miyasaka (Osaka University). BC-1 cells were seeded into 24-well plates at 50,000 cells/well and treated with L-(−)-norepinephrine-(+)-bitartrate (10, 100 and 300 μM; EMD Millipore, Billerica, MA) in Dulbecco’s Modified Eagle Medium (supplemented with 10% FBS; Gibco by Life Technologies) for 48 h after 2 h serum starvation. Cells were then dissociated with 0.05% Trypsin/0.53 mM EDTA (Nacalai Tesque, Kyoto, Japan), pelleted and suspended in RP10 medium, and immediately used for flow cytometry analysis.

Adrenergic Receptor Assay Protocol

(±)-epinephrine HCl, (L)-norepinephrine bitartrate, (R)-phenylephrine HCl, sodium pentobarbital, Nile Red dye and solvents for drug delivery were all obtained from Sigma (St. Louis, MO). [3H] norepinephrine (13.9 Ci/mmol) and Ultima Gold liquid scintillation cocktail were obtained from Perkin Elmer (Boston MA). Proteinase K (# 03 115887001) was from Roche. Female Golden Syrian hamsters (100 g) and ICR female mice (20–25 g) were purchased from Harlan Laboratories (Indianapolis, IN). Mice and hamsters were maintained on a 12 h light/dark cycle and provided water and Harlan 5305 (mice) or Harlan 2018 (hamsters) lab chow ad libitum. Animal procedures were approved by University of Wisconsin IACUC (Protocol # M0476).

Maternal exposure effects on neonatal rats

The PrC-210 aminothiol was synthesized as described previously.18 Lactating female Sprague–Dawley rats with litters of 10-day-old pups were obtained from Harlan (Indianapolis, IN). Rats were maintained on 12-hr light/dark cycle and provided ad lib water and lab chow. All animal procedures were conducted according to a protocol (#M0476) approved by the University of Wisconsin Institutional Animal Care and Use Committee.
