Preparing DON Standards for Microbial Analysis

DON/3-keto-DON standards were purchased from TripleBond (Guelph, Canada), solubilized in sterile water to bring the initial concentrations to 1.0 mg/ml, filtered with 0.22 μm membrane filters, and stored at 4°C until used. Water was purified by a Milli-Q water system (Millipore Corporation, New York, USA). Mineral salt medium (MM) was formulated as described previously (Ikunaga et al., 2011 (link)), but with slight modifications. In brief, the medium contained 1.6 g of Na2SO4, 1.0 g of KH2PO4, 0.5 g of MgSO4, 0.5 g of NaNO3, 0.5 g of (NH4)2SO4, 0.025 g of CaCl2, and DON as the carbon source. Mineral salt-based medium (MSB) was composed of basic MM supplemented with 10% peptone. Luria-Bertani (LB) medium contained 10.0 g of tryptone, 5.0 g of yeast extract, and 10.0 g of NaCl. HPLC-grade methanol and analytical-grade ethyl acetate were obtained from Sinopharm (Shanghai, China). Cycloheximide was purchased from Solarbio (Beijing, China).

Tripterine-Based Lipid Nanoformulation for Targeted Drug Delivery

Tripterine was obtained from DESITE Biotechnology Co., Ltd (Nanjing, China). HPLC Analysis of β-asarone in Brain Tissue

Measurement of β-asarone in brain tissues was performed by High Performance Liquid Chromatography (HPLC) analysis. 1g of brain tissues were grinded in mortar with 1ml of cold phosphate-buffered saline (pH 7.4). The brain tissue was transferred into a tube to homogenize by sonication (50W×15s) on ice. β-asarone was extracted by 1.5ml HPLC-grade hexane (Sinopharm Chemical Reagent Co., Ltd), and then the organic layer was filtered. β-asarone was separated under isocratic condition using 70% HPLC-grade methanol (Sinopharm Chemical Reagent Co., Ltd) in ultrapure water and a column (Inertsil ODS-3, 5μm, 4×250mm). The absorbance of the organic layer was measured at 257nm. Compound identification and analysis calibration were based on use of β-asarone as external standards.

Synthesis and Characterization of Polymeric Sorbents

Both bisultap and monosultap with purities over 99% were from Crescent Chemical Company, Islandia, NY, USA. Triple distilled water (deionized water) newly collected from a glass distillator (Yarong, Shanghai, China) was used to prepare solutions. Other reagents were of analytical grade and obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). Six tea samples (green, black, white, yellow, dark and oolong teas), bottled green tea beverage, and bottled iced black tea beverage were purchased from a local supermarket.

Simultaneous Quantification of Endocrine Disruptors

All reagents were at least analytical reagent-grade and were used as received. BPA, BPF, OP and NP were purchased from Sigma (Shanghai, China). HPLC-grade acetonitrile and HPLC-grade mEthanol were obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). 1,3,5-Triformylphloroglucinol (Tp) was purchased from Aladdin (Shanghai, China), and benzidine (BD) was purchased from J&K (Beijing, China). Ethanol and N,N-dimethylformamide (DMF) were obtained from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). Formic acid and acetic acid were purchased from Aladdin (Shanghai, China). Ultrapure water (18.2 MΩ cm−1) was generated by the Sartorius Milli-Q system (Aubagne, France) and was used throughout the experiments. All the glassware was cleaned with diluted HNO3 (10%) and thoroughly rinsed with ultrapure water prior to use. Mixed standard stock solutions (1000 mg L−1) were prepared by dissolving appropriate amounts of four targets in acetonitrile and storing in dark at 4 °C. Working solutions were obtained daily by diluting the stock solutions with ultrapure water before experiments.

Apoptosis and ROS Detection in P. baumii

Microbial Cultivation Medium Preparation

Atrazine Extraction and Quantification in Soil

Extraction of atrazine from soil samples was performed in the manner described by Krutz et al. [17 (link)]. The extracts were analyzed by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) on an UltiMate 3000 HPLC system interfaced to a TSQ Vantage Triple Quadrupole Mass Spectrometer (Thermo Fisher Scientific, USA). The conditions of HPLC and MS/MS parameters are provided in Additional file 12. Calibration standards of atrazine (# 45330 Sigma-Aldrich, USA) were prepared in HPLC grade methanol (Sinopharm, China) at the following concentrations: 0.01, 0.05, 0.1, 0.5, 1, 5, 10, 50 and 100 ng mL−1. The typical calibration curve was Y = 2313.0 + 4073.8X. The curve displayed excellent linearity (r2 = 0.9995) over the entire calibration range. Recovery of atrazine from fortified soil samples was 90.3–93.2 %. The method limit of atrazine quantification was 0.05 μg kg−1.
