Echocardiographic Evaluation of Rat Heart

Angiotensin II-Induced Aortic Aneurysm

Male ApoE -/-mice (10-18 weeks) were obtained from the Jackson laboratory (B6.129P2-Apoe tm1Unc /J), fed a standard laboratory chow and randomly assigned to the AngII or control group. Alzet osmotic mini pumps (Model 1004) were implanted to deliver 1000 ng/kg/min of Angiotensin II (Sigma-Aldrich) for a period of 14 or 28 days. Three days before implantation a western diet (1.25% Cholesterol, ssniff) was started. Disease progression was monitored using a Vevo 3100 ultrasound machine (Visual Sonics, Toronto, Canada). As controls, ApoE -/-animals subjected to sham surgeries and western diet were used. All procedures were approved by the local government authorities (LANUV, NRW, Germany). In accordance with earlier studies from other groups we have used males mice, as akin to human males, they are more susceptible to aneurysm formation [13] (link). In addition, when focusing on fundamental pathogenic processes the standardized use of males helps to overcome biases, to enhance reproducibility and comparability across aneurysm studies [14] (link).

Abdominal Aorta Pulse Wave Velocity

At the end of the experiment, pulse wave velocity (PWV) in mice from each group were detected by Vevo3100 Ultrasound Machine (FUJIFILM VisualSonics, Canada) as reported previously (Wu et al., 2021 (link)). Briefly, mice with abdominal hair removed were anesthetized with 2.5% isoflurane and maintained with 1.5% isoflurane and fixed in a 37°C pre-heated platform with a heart rate of 400–550 bp. MX400 (30 MHz) probes for mice were placed longitudinally below the sternum and xiphoid process to get the full image of the abdominal aorta in both B mode and M mode. The PWV was calculated by the formula: length of abdominal aorta (distal time delay-proximal time delay). Images were taken in both B/M mode, followed by the PWV via Vevo® LAB software (FUJIFILM VisualSonics, Canada).

Cardiac Function Assessment in Mouse MI Model

Systolic blood pressure and heart rate were non-invasively and measured in the awake state using a tail cuff method with a MK-2000 (Muromachi Kikai, Tokyo, Japan). On the indicated days before and after MI, transthoracic ultrasound cardiography was performed using a Vevo3100 ultrasound machine (FUJIFILM VisualSonics, Toronto, Canada) as described previously [17 (link), 21 (link)]. Mice were kept under light anesthesia using isoflurane (5% for induction; 1–2% for maintenance) with the heart rate over 450 bpm. Left ventricular internal dimension in diastole (LVID:d), left ventricular internal dimension in systole (LVID:s), interventricular septal thickness in diastole (IVS:d), and left ventricular posterior wall thickness in diastole (LVPW:d) were measured in M-mode at the level of the papillary muscle.
