Iron chloride and Tween 80 were purchased from Tianjin Guangfu Fine Chemical Research Institute (Tianjin, China). Ethylene glycol, trisodium citrate, and sodium acetate were purchased from Beijing Chemical Works (Beijing, China). Acrylic acid (AA, purity ≥ 98%) was supplied by Wulian Chemical Factory (Shanghai, China). Potassium persulfate was purchased from Sinopharm Chemical Reagent Co., Ltd. (Beijing, China). N, N′-methylenebisacrylamide was purchased from Junyao Albert Biotechnology Company (Beijing, China). Liquid paraffin was purchased from Tianjin Bodi Chemical Co., Ltd. (Tianjin, China). Span 80 was purchased from Xilong Chemical Co., Ltd. (Shantou, China). Hydroxylamine hydrochloride and O-phenanthroline Hydrate were obtained from Yingdaxigui Chemical Reagent Factory (Tianjin, China). Agarose, pentobarbital sodium, and sodium carboxylmethyl cellulose were purchased from Sigma Aldrich (USA). Embospheres were purchased from Biosphere Medical Inc. (USA). Iodixanol injections were supplied by GE Healthcare Ireland (Ireland). All the other solvents and reagents were of analytical grade and used as received without further purification otherwise noted.

Fag Extraction and Preparation

Fag was extracted twice (100 g in 800 ml 50% ethanol) by conventional refluxing for 2 h. The extracts were combined, filtered and concentrated under reduced pressure in a vacuum at 60°C to form a residue (9.48 g). The residue was resolved in 0.5% sodium carboxyl methylcellulose (Sigma-Aldrich; Merck KGaA) for animal experiments and in vitro testing.

Animal Handling and Reagent Procurement for Mouse Studies

Six-week-old ICR, Balb/c, and C57BL/6 male mice (see Table S2 for genetic background information) were purchased from Daehan Biolink (DBL, Chungbuk, Korea) and housed eight mice per group under a 12-h light/dark cycle (lights on at 6 a.m.). Water and a pellet diet (Samyang, Daejeon, Korea) were supplied ad libitum. Animal care followed guidelines issued by the National Institutes of Health for the Care and Use of Laboratory Animals (NIH Publication 80-23, revised in 1996) and the Institutional Animal Care and Use Committee at Sungkyunkwan University (Approval No.: SKKUIACUC-2016-01-0002-2). Phorbol-12-myristate, sodium carboxyl methylcellulose, 2,4-dinitrochlorobenzene (DNCB), dextran sodium sulphate (DSS), acetylsalicylic acid, D-GalN, LPS (E. coli 0111:B4), 100% EtOH, and HCl were purchased from Sigma Chemical Co. (St. Louis, MO, USA). MAPK inhibitors (SB203580, SP600125, and U0126) were purchased from Calbiochem (La Jolla, CA, USA). RAW264.7, HEK293, and MDA-MB-231 cells were purchased from the American Type Culture Collection (Manassas, VA, USA). Detailed information on antibodies used in this study is explained in Supplementary Materials. The AP-1 luciferase construct was purchased from Addgene (Cambridge, MA, USA).

Establishment and Characterization of Luciferase-Tagged Human Liver Cancer Cell Lines

The human HCC cell line MHCC97L, which was tagged with a luciferase reporter gene, was gifted by Professor Man Kwan from the Department of Surgery, The University of Hong Kong. The HLF cell line was purchased from Japanese Collection of Research Bioresources cell bank. Both cell lines were cultured in Dulbecco's modified Eagle's medium (Gibco, Waltham, MA, USA) supplemented with 4.5g/L glucose, 10% foetal bovine serum (Gibco, MA, USA) and 1% penicillin/streptomycin (Gibco, MA, USA)); and they were kept in a 37 °C humidified incubator (Thermo Fisher Scientific Inc., Waltham, MA, USA) under 5% CO 2 . The DNA plasmid encoding pBABE-puro LEF1 was gifted by Joan Massague (plasmid #27023; Addgene Inc., Cambridge, MA); and the DNA plasmid expressing pAd/WT1-IRES-nAmCyan was gifted by Edward McCabe (plasmid #29756, Addgene). Mangiferin was purchased from Sigma-Aldrich Co. (St. Louis, MO, USA). The stock solution of Mangiferin (100 mg/mL) was prepared in DMSO (Sigma-Aldrich Co., St. Louis, M.O., USA) and dH 2 O (1:9, v/v). For animal administration, Mangiferin (50 mg/kg) was prepared in 0.4% of sodium carboxyl methyl cellulose (Sigma-Aldrich Co., St. Louis, M.O., USA).
