The cells were seeded in 12-well plates and infected with H1N1/UI182 (MOI = 0.5) when confluence reached 70%. At 48 h post-infection, cells were washed three times with cold PBS and fixed with 4% paraformaldehyde (PFA) for 20 min, then the plate was infiltrated with 0.2% Triton-X100 and blocked with 2% BSA for 1 h. The primary antibody against influenza virus Nucleocapsid protein (Abcam, ab104870, 1:200) was added and incubated with cells overnight at 4 ℃.
