Knockdown of Circadian Clock Gene Per1 in SCN

4-AP, TTX, cadmium chloride, and IbTX were purchased from Sigma-Aldrich (St. Louis, MO). Gabazine (SR 95531 hydrobromide) was purchased from Tocris (Bristol, UK). For Per1 gene knock down, either antisense ODN sequence against the 5′ translational start site of Per1 (taggggaccactcatgtct; Sigma-Aldrich) or a random ODN with equivalent GC content (ccgttagtactgagctgac) was applied at a final concentration of 10 µM from ZT 2 to ZT 4. These antisense ODN and random ODN were previously used (Akiyama et al., 1999 (link); Wakamatsu et al., 2001 (link); Tischkau et al., 2003 (link); Marquez et al., 2004 (link); Koyanagi et al., 2006 (link); Gamble et al., 2007 (link); Kudo et al., 2013 (link)). While siRNA can be a more efficient method, there are problems of transfection efficiency while the ODN method to reduce Per1 has been established in the SCN slices (Gamble et al., 2007 (link); Kudo et al., 2013 (link)). The antisense ODN is incorporated into cells by endocytosis. The antisense ODN was applied from ZT 2 to ZT 4, because the SCN slices was moved from 35-mm culture dish to the perfusion chamber. In the perfusion chamber, the flow rate is 2 ml min−1 and it is cost prohibitive to continually apply antisense ODN through the perfusion system.

Nicotine, D2 Receptor Modulation in Neurons

Quinpirole hydrochloride, eticlopride hydrochloride, and gabazine (SR 95531 hydrobromide) were purchased from Tocris Bioscience. (-)-Nicotine hydrogen tartrate salt was purchased from Sigma-Aldrich. 15 min after the first nicotine injection, quinpirole 1 mg/kg was administered intravenously. 5 minutes later, eticlopride 1 mg/kg was administered intravenously. When the neuron activity was returned to baseline, a second nicotine injection was performed. Once D2-R pharmacology was applied, no further neurons were recorded and the animal was discarded.

Pharmacological Modulators of Neuronal Signaling

Bumetanide, VU0240551, picrotoxin, nimodipine, and GABAzine (SR 95531 hydrobromide) were acquired from Tocris Bioscience (Bristol, United Kingdom). Bumetanide and picrotoxin were dissolved and stored in DMSO at 100 mM. VU0240551 was dissolved in DMSO at 20 mM. GABAzine stock was dissolved in water at 2 mM. The anion exchange blocker 4,4’-Diisothiocyano-2,2’-stilbenedisulfonic acid (DIDS) was acquired from Sigma-Aldrich (St. Louis, MO) and stored in DMSO at 100 mM before use. Strychnine hydrochloride, diazepam, zolpidem, and 5-Nitro-2-(3-phenylpropylamino)benzoic acid (NPPB) were purchased from Sigma-Aldrich (St. Louis, MO).

Drug-containing stock solutions were diluted to desired concentrations in either ACSF. All drugs were applied via the “Y-tube” application adapted to brain slices (Murase et al., 1989 (link); Hevers and Lüddens, 2002 (link)).
