Fetal Motor Neuron Transplantation in Rats

The animal rearing management was carried out according to the Kitasato University Faculty of Veterinary Medicine Animal Experiment Guideline and Manual for Rearing and Management of Experimental Animals (Approval No: 17-127, 18-127, 19-085). The rats were housed in cages under temperature and light-controlled conditions in a 12-hour cycle and were provided with fresh food and water ad libitum.
In Experiment 1, we used three pregnant female Lewis rats on gestation day 15 (E15) (Japan Charles River Laboratories, Kanagawa, Japan) to obtain fetal MNB. As recipient rats (organ recipient animals), we used 12 male Lewis rats (Japan Charles River Laboratories) aged 11 weeks, with a body weight of 309.0 ± 11.4 g.
In Experiment 2, we used three pregnant female Lewis rats on E15 (Japan Charles River Laboratories), and as recipient rats, 18 male Lewis rats (Japan Charles River Laboratories) aged 9 weeks, with a body weight of 243.0 ± 7.5 g.
In Experiment 3, we used three pregnant female Lewis rats on E15 (Japan Charles River Laboratories), and as recipient rats, 9 male Lewis rats (Japan Charles River Laboratories), aged 10 weeks, with a body weight of 292.8 ± 7.6 g.

Rheumatoid Arthritis Rat Model Establishment and Analysis

Male Lewis rats (n = 48, 6–8-week-old, 200 ± 20 g in weight) were purchased from Beijing Vital River Laboratory Animal Technology Co., Ltd. (production license no. SCXK 2016-0006, Beijing, China). All animals were maintained under specific pathogen-free conditions in a temperature-controlled room at a constant temperature of 24 ± 1 °C with a 12 h light/dark cycle and had free access to the standard rodent diet and water ad libitum. The study was approved by the Research Ethics Committee of the Institute of Basic Theory of Chinese Medicine, China Academy of Chinese Medical Sciences, Beijing, China (SYXK 2016-0021, Beijing, China). All animal studies were treated in accordance with the guidelines and regulations for the use and care of animals of the Center for Laboratory Animal Care, China Academy of Chinese Medical Sciences.
A total of 48 rats were randomly divided into three groups: ① normal control group (n = 16); ② AIA-Cold (RA-Cold model group, n = 16); ③ AIA-Hot (RA-Hot model group, n = 16). Among them, AIA-Cold and AIA-Hot rat models were established and evaluated according to our previous studies [9 (link), 11 (link)]. Urine and serum samples were collected 28th d after Freund complete adjuvant stimulation, after centrifugation at 3000 rpm for 15 min, the supernatant was reserved and stored at − 80 °C for subsequent metabonomics analysis.

Experimental Autoimmune Myocarditis in Rats

Male Lewis rats (180‐200 g), aged 6‐8 weeks, purchased from Beijing Vital River Laboratory Animal Technology (Beijing, China) were maintained in Xiamen University animal experimental centre. All animal care and experiments were performed in accordance with procedures approved by the Animal Care and Use Committee of Xiamen University. Porcine cardiac myosin was prepared from the ventricular muscle of porcine hearts as previously described.29 To produce EAM, each rat was immunized on day 0 with 0.2 mL emulsion containing 1 mg porcine cardiac myosin with an equal volume of complete Freund's adjuvant supplemented with mycobacterium tuberculosis H37RA at a concentration of 10 mg/mL by a single subcutaneous injection in both footpads. The rats in the control group were only immunized with complete Freund's adjuvant.

Xenogeneic Bone Marrow Mesenchymal Stem Cell Transplantation

Inbred adult male Brown Norway (BN) rats (180–200 g) and male Lewis rats (200–220 g for SBTx and 100–120 g for BMMSCs) were obtained from Vital River Company (Beijing, China). Animals were housed individually in standard animal facilities, maintained on a 12-h light/dark cycle and provided with commercially available chow and tap water ad libitum for 3 days before testing. Food was withheld from both donor and recipient animals for 24 h prior to surgery. All experimental procedures were carried out in accordance with the Guide for the Care and Use of Laboratory Animals published by the National Institutes of Health (NIH publication 86–23, revised 1985), and the protocols were approved by the Animal Care and Research Committee of Tianjin First Central Hospital, Tianjin, China (Permit Number: E20130605-001A). All surgery was performed under chloral hydrate anesthesia, and all efforts were made to minimize animal suffering.

Liver Transplantation in Rat Model

Specific-pathogen-free healthy adult inbred male Brown-Norway (BN) rats (n=45) and male Lewis rats (n=35) were purchased from the Vital River Laboratories Animal Technology, Co., Ltd. (Beijing, China). The rats were housed individually in standard animal facilities at 18–26°C with a 12-h light/dark cycle and were provided with commercially available chow and tap water ad libitum. BM-MSCs were extracted from syngeneic inbred male BN rats (80–100 g; 4–5 weeks old). Inbred male Lewis rats (210–250 g; 8–10 weeks old) were the liver transplantation donors, and inbred male BN rats (210–250 g; 8–10 weeks old) were the recipients. The difference in body weight between the donor and the recipient of each pair did not exceed 10 g. Food was withheld from the recipient animals for 12 h before surgery, but was not withheld from the donor animals. The study protocol was approved by the Animal Care and Research Committee of Tianjin First Central Hospital (Tianjin, China). All surgeries and sacrifices were performed under 5% chloral hydrate anesthesia (0.5 ml/100 g). Every effort was made to minimize animal suffering.

Establishing Rat Model for Research

Animal Housing and Acclimatization
