Probiotic S. boulardii-YS01 Cultivation

NMN (>95%), NR (≥95%), Nam (≥99.5%), and NA (≥99%) were purchased from Shanghai yuanye Bio-Technology Co., Ltd. (Shanghai, China); NAD+ (>98%) was purchased from Shanghai Macklin Biochemical Co., Ltd. (Shanghai, China). Ultra-pure water was prepared using a Milli-Q water system (Milli-pore–Waters, Milford, MA, USA). HPLC-grade methanol was obtained from Fisher Chemicals (Fairlawn, NJ, USA). The complex medium of S. boulardii-YS01 contained (g/L) glucose (10), saccharose (20), peptone (10), yeast extract (10), malt powder (1), Na2CO3 (0.3), KH2PO4 (1.5), MgSO4 (1), ammonium citrate tribasic (5), ammonium ferric citrate (0.02), glycerin (10), and multivitamin B tablet (0.1 piece/L). All of them were dissolved in deionized water and then filled to their final volume. The corresponding solid medium was prepared by adding 2% AGAR powder to the liquid medium. The media and vessels (e.g., Erlenmeyer flask and centrifuge tubes) were sterilized in an autoclave at 115 °C for 30 min.

Comprehensive Ginseng Root Profiling

Thirty-six batches of representative cultivated ginseng root were collected or purchased from different cultivation areas in Jilin, Liaoning, and Heilongjiang Provinces, the main source of ginseng in China. A detailed sample list is provided in Table S2. The identity of all samples was confirmed by Prof. Bao-lin Guo of the Institute of Medicinal Plant Development, Beijing, China. Each whole ginseng root was divided into four parts: main root (MR), branch root (BR), rhizome (RH), and fibrous root (FR). One hundred forty-four different parts samples of ginseng root are present.
Ginsenosides Rb1, Re, Rg1, Rd, Rc, Rf, Ro, Rg2, 20(R)-Rh1, 20(S)-Rh2, 20(R)-Rh2, 20(S)-Rg3, and 20(R)-Rg3 and notoginsenosides R1 and R2 were isolated in our laboratory and identified by spectroscopic data. The purities of these standards were better than 98% by the HPLC analysis. All samples were stored at 4 °C before use.
Acetonitrile (HPLC grade) was purchased from Fisher Scientific Co. (Loughborough, UK). Distilled water was purchased from Watsons (Guangzhou, China). Phytochemical Analysis of Polygonum capitatum
