Cells were seeded on a 6-well plate at a density of 3 × 105 cells/well in 1 ml of complete medium in quadruplicate with relevant siRNA transfections and chemical inhibitor treatments. After a 48 h or 72 h incubation, the supernatants (i.e., conditioned medium) were collected and spun down at 3000g for 10 min to remove any cell debris.
