In Vitro Rumen Fermentation with Melatonin

The in vitro simulated fermentation solutions were shown in Table 5. A total of three different treatments were included: (i) non-rumen fluid + non-melatonin (CK0); (ii) rumen fluid + non-melatonin (CK1); (iii) rumen fluid + melatonin (MLT). Three replicates for each treatment. The 1.2 g feed provided by Youran Dairy Farm (Inner Mongolia, China; Table 4) was weighed and used as fermentation substrates for every fermentation bottle. The standard solutions of MLT (9 ng/mL) were prepared according to Özcan and Bagci (61 (link)) using melatonin powder purchased from Sigma-Aldrich company (Sigma-Aldrich, USA). The rumen fluid was collected from four different directions of rumen of three fistula healthy Holstein cows after 2 h of morning feeding. Artificial saliva in vitro culture experiments were prepared according to Cone et al. (62 (link)). All incubation procedures were performed in a thermostatic water bath shaker (SHA-A, Hengfeng Instrument, China) at 39°C (63 (link)). After 24 h of cultivation, the fermentation bottle was taken out and placed in an ice water bath to stop the fermentation.

Melatonin's Effects on Microcirculation

To study the effects of melatonin, in total 3.3 mg melatonin [melatonin powder, Sigma-Aldrich, St. Louis, MO, USA; 0.3 mg melatonin diluted in NaCl 0.9% to a total volume of 3 ml, 5 min later followed by 3 mg melatonin diluted in 3 ml NaCl 0.9% containing 5% ethanol (Carl Roth, Karlsruhe, Germany)] was applied to the oral and gastric mucosa, and all variables were recorded for 2.5 h. melatonin was applied next to the site of the microcirculation measurement. The gastric bolus reached the stomach via a gastric tube fixed to the O2C probe, whereas the bolus to the oral mucosa was applied around the measuring point via a perforated silicon tube. As melatonin is poorly soluble in water, we choose the solvent ethanol. melatonin mediated effects on local microcirculation may be superseded by direct effects of ethanol. Therefore, melatonin was applied in two subsequent dosages with the lower concentration dissolved in NaCl 0.9% and the higher dissolved in ethanol. To assess early effects of melatonin on local microcirculation, microcirculatory oxygenation and perfusion were analyzed 5 min after each melatonin application.

Melatonin Effects on Sirohi Buck Semen

The bucks were randomly divided into 2 groups with 6 animals in each group for the present experiment in order to study effect of exogenous melatonin administration on freezability of Sirohi buck semen during non-breeding season in Southern Rajasthan. The first group treated (T) animals (n=06) were administered single subcutaneous injection of melatonin (Melatonin powder Sigma, USA dissolved in sterile corn oil) @18mg/50kg body weight as per previously described methods (Chemineau et al., 1995, Kumar and Purohit, 2009, Kumar, 2014) [3, 17, 18] .The second group of bucks (n=6) were given only corn oil subcutaneously and considered as control group (C). The semen collections were done aseptically and hygienically on weekly basis during nonbreeding seasons. The artificial vagina (Danish model) of size 20 cm in length and 4.5 cm in diameter with smooth lining (IMV Pvt. Ltd.) was used for semen collection. The semen collection cup with the freshly collected semen was immediately transferred to the laboratory and immersed in a water bath at 30 ºC. Total 48 semen samples non-breeding season-I and 96 semen samples non-breeding season-II during 4 weeks and 8 weeks were collected from the control (n=6) and treated (n=6) buck group per week in the semen collection area of semen laboratory, in a clean dust-free area.
