Determination of PUFA Composition in Oviductus Ranae

Petroleum ether, methanol, and phosphoric acid were analytical pure and purchased from Beijing Chemical Factory (Beijing, China). HPLC-grade acetonitrile and HPLC-grade methanol were purchased from Fisher (Fisher Scientific, Fair Lawn, NJ, USA). The ultrapure water was prepared from a gradient water purification system (Water Purifier, Sichuan, China). Eicosapentaenoic acid (EPA, Lot No: 32510020), docosahexaenoic acid (DHA, Lot No: 0501808), and arachidonic acid (ARA, Lot No: 2491801) were purchased from TanMo Quality Testing Technology Co., Ltd. (Beijing, China). α-Linolenic acid (ALA, Lot No: U-62A-J25-C), linoleic acid (LA, Lot No: U-52A-JA3-C), and oleic acid (OA, Lot No: U-46A-MA6-C) were purchased from ANPEL Laboratory Technologies (Shanghai) Inc. (Shanghai, China). The purity of each standard compound was higher than 99%, as determined by HPLC. Six kinds of PUFAs standards were formulated to different concentrations using HPLC-grade methanol and filtrated with a 0.22 μm filter membrane prior HPLC injection.
Jilin Province Rana Industry Association provided a total of 27 samples of Oviductus Ranae, which were collected from the Changbai Mountains area in Jilin Province, China. Collection sources covered the major production areas of Changbai Mountains. The specific collection information is shown in Table 1, and Figure 2 shows the geographical distribution of the samples.

Analytical Methods for Soybean Oil and Potato Chips

Quick‐frozen potato chips were supplied by Shanghai Sheng Fang Industrial Company, Limited. Sodium hydroxide, ether (absolute), isopropyl alcohol, ethanol, trichloroacetic acid, benzene, potassium hydroxide, and petroleum ether were obtained from Beijing Chemical Works. Phenolphthalein was provided by Tianjin Beichen Fangzheng Reagent Factory. 2,4‐Dinitrophenylhydrazine was obtained from Tianjin Ruijin Chemical Company, Limited.
HH‐S digital display constant temperature oil bath was supplied by Changzhou Rong Hua Instrument Manufacturing Company, Limited. HH‐ZK1 constant temperature control was obtained from Gongyi Yuhua Instrument Liability Company, Limited. The spherical condensation tube and glass dryer were supplied by Beijing Huabo Station Biological Analysis Technology Company, Limited. UV‐9000S double beam UV–vis spectrophotometer was obtained from Shanghai Yuanshan Instrument Company, Limited. The YZF vacuum dryer was purchased from Shanghai Yao Instrument Equipment Factory.

NMR and MS Analysis of Organic Compounds

NMR spectra were recorded on a Bruker Avance III 600 spectrometer (Bruker, Billerica, Germany) with 1H-NMR at 600 MHz and 13C-NMR at 150 MHz, with chemical shift values expressed as δ values, using deuterated solvent signals as an internal reference. HR-ESI-MS was performed using an LTQ-Orbitrap XL spectrometer (Thermo Fisher Scientific, Boston, MA, USA). Acchrom S6000 high-performance liquid chromatograph (Acchrom Tech Technology Co., Ltd., Beijing, China) was used for analysis with an ELSD-UM 5800 Plus (Unimicro Technologies Co., Ltd., Shanghai, China). Column chromatography (CC) analysis was performed using silica gel (200–300 mesh and 300–400 mesh, Qingdao Ocean Chemical Factory, Qingdao, China) and C18 reverse silica gel packing (50 μm, YMC Co., Ltd., Tokyo, Japan). Analytically pure solvents (petroleum ether, ethyl acetate, ethanol, methanol, n-BuOH, and dichloromethane) (Beijing Chemical Factory, Beijing, China) were used for extraction and CC separation. Chromatographically pure methanol (Thermo Fisher Scientific, Waltham, MA, USA) was used for high-performance liquid phase analysis. Deuterated solvents (Deuterated methanol, deuterated pyridine, deuterated chloroform, and deuterated dimethyl sulfoxide) (Aladdin Biochemical Technology Co., Ltd., Shanghai, China) were used for nuclear magnetic resonance spectroscopy.

Ferrous chloride, sodium salicylate, ferrous sulphate, hydrogen peroxide, potassium ferricyanide, trichloroacetic acid, disodium phosphate, sodium dihydrogen phosphate, ferric chloride, sodium carbonate, sodium hydroxide, aluminium nitrate, sodium nitrite, petroleum ether (60–90 °C), ethanol and methanol were all purchased from Beijing chemical plant (Beijing, China). All chemicals were of analytical grade. Water was treated in a Milli-Q water purification system (TGI Pure Water Systems, USA).

Synthesis and Characterization of Fluorinated Monomers

Isolation and Purification of Ranae Oviductus

The petroleum ether, guanidine hydrochloride, and ammonium sulfate analytical grade were purchased from Beijing Chemical Factory (Beijing, China). The dithiothreitol (DTT) and trifluoroacetic acid (TFA) were purchased from Sigma-Aldrich (St. Louis, MO, USA). The HPLC-grade acetonitrile (MeCN) and HPLC-grade methanol were purchased from Fisher (Fisher Scientific, USA). The ultrapure water was obtained from a gradient water purification system (Water Purifier, Sichuan, China).
Ranae Oviductus, bullfrog oviduct, toad oviduct and frog oviduct were provided by Jilin Province Rana Industry Association which were collected from the Changbai Mountain area in the Jilin province of China. The specific location is shown on the map in Figure 1. Ten batches of Ranae Oviductus samples were collected from different regions from the main producing area of the Changbai Mountain range. The specific collection information is shown in Table 1.
