Modulating Astrocyte Responses to S100B

TLR7 and Chloroquine Modulate Inflammatory Cytokines

TLR7 siRNA plasmid (sc-40266, Santa Cruz Biotechnology, Inc, California, USA) and control siRNA plasmid were from Santa Cruz Biotechnology. Interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) were evaluated by commercially available enzyme-linked immunosorbent assay (ELISA) kits according to the manufacturer's protocol (Elabscience Biotechnology, Wuhan, China). Chloroquine was dissolved in calcium-and magnesium-free phosphate-buffered saline at a concentration of 10 mM. Stock solutions were diluted to the desired final concentrations with growth medium just before use.
