Murine Bone Marrow-Derived Macrophage Isolation

To prepare bone marrow-derived macrophages (BMDMs), bone marrow cells from WT and Tyk2 KO mice were cultured in RPMI supplemented with 10% FBS and recombinant murine macrophage colony stimulating factor (100 ng/ml; PeproTech, Rocky Hill, NJ) for 6 days. BMDMs and peritoneal resident cells collected from the peritoneal lavage were plated in RPMI-1640 medium supplemented with 10% FBS, 100 U/ml penicillin and 0.1 mg/ml streptomycin. IL-10 levels in the culture supernatants of BMDMs or peritoneal resident cells were determined using an IL-10 ELISA kit (eBioscience). The following agents were tested for their effects on IL-10 production: diclofenac sodium (10 µM; Wako Pure Chemical Industries, Osaka, Japan), and H-89 (10 µM; Cell Signaling Technologies, Beverly, MA).

Synoviocyte Metabolism Modulation

Glucosamine hydrochloride, D-[6-3H (N)] ([3H]glucosamine) (37 MBq/mL) and Ultima-FloTMM (scintillation solution) were from PerkinElmer Co., Ltd. (Waltham, MA). OH pak SB-805 HQ and OH pak SB-807 HQ columns were from SHOWA DENKO K.K. (Tokyo, Japan). Select-HATM 500 k, Select-HATM 1000 k, and Select-HATM 2500 k were from Hyalose, LLC. (Oklahoma City, OK).

Fabrication of Microneedle Patch with Diclofenac

Sodium hyaluronate (HA) (FCH-SU, lot: HGV016900, average molecular weight (MW): 50,000-110,000 Da) was purchased from Kikkoman Biochemifa Company (Tokyo, Japan). Diclofenac sodium (DCF) was purchased from FUJIFILM Wako Pure Chemical Corporation (Osaka, Japan).
Polydimethylsiloxane (PDMS, Dowsil Silpot 184) was purchased from Dow Corning (Midland, MI, USA). Aluminum microneedle mold masters, fabricated via micro-milling, were purchased from Tokai Azumi Techno (Mie, Japan). All other chemicals were purchased commercially as reagent-grade products.
