Rosmarinic Acid and SalB Nanoparticles

Rosmarinic acid (99.8%) and SalB (99.5%) were purchased from the National Institute for the Control of Pharmaceutical and Biological Products (NICPBP, Beijing, China). Water-soluble CS with various MWs (3, 30, 50, and 100 kDa) were supplied by KITTOLIFE (Seoul, Korea). Cloned Caco-2 TC7 cells were a generous gift from Dr. Ming Hu of INSERM U178 (Houston, TX). Sodium TPP was purchased from Merck (Darmstadt, Germany). 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) and Hank's balanced salt solution (HBSS; powder form) were purchased from Sigma-Aldrich (St. Louis, MO). Milli-Q water (Millipore, Bedford, MA) was used throughout the study. All the reagents were of analytical grade, except acetonitrile and methanol, which were chromatographic grade (Merck Company Inc., UN).

Optimized Irinotecan Formulation Preparation

irinotecan HCl Trihydrate injection (20 mg/mL) were purchased from Teva Parenteral Medicine, Inc. (Irvine, NY, USA) and the powder of irinotecan were purchased from Dalian Meilun Biology Technology Co., Ltd. (Dalian, China), respectively. The self-made injectable irinotecan formulation was prepared as follows: the irinotecan hydrochloride was firstly dissolved in 1,3 Propanedio by sonication 10 min, then diluted in a sorbitol/lactic acid buffer (D-sorbitol 45 mg/ml and D-lactic acid 0.9 mg/ml, pH=3.4), the control group was given 0.2 mL sorbitol/lactic acid buffer.
The reference standard irinotecan, SN-38, SN-38G, NPC, and APC were purchased from Toronto Research Chemicals Inc. (Toronto Canada). Uridine-5’- diphosphate-β, D-glucuronic acid ester, D-saccharic-1, 4-lactone monohydrate, magnesium chloride, daidzin, and astragaloside were purchased from Sigma–Aldrich Co. (St. Louis, MO, USA). Acetonitrile, methanol, and formic acid (LC-MS grade) were purchased from VWR. Glucose, Hank’s balanced salt solution (HBSS, powder form), HEPES, Ko143, MK-571, Zosuquidar (LY335979), NaHCO3, and fetal bovine serum were purchased from Sigma-Aldrich Co., (St. Louis, MO, USA). Transwell™ cell culture insert (polycarbonate membrane and polystyrene plates with a diameter of 24 mm and a pore size of 3 μm) was obtained from Corning Incorporated (Corning, NY, USA).

Caco-2 Permeability Assay Protocol

Transwell permeable polycarbonate inserts (0.4 μm) and 12-well cell culture plates were obtained from Corning. (Cambridge, MA, USA). The Caco-2 cell line was generously provided by the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). Dulbecco’s modified Eagle’s medium (DMEM) was obtained from Gibco Laboratories (Life Technologies Inc., Grand Island, NY, USA). Hanks’ balanced salt solution (HBSS, powder form) was obtained from Sigma Chemical Co. (Deisenhofen, Germany). Fetal bovine serum was purchased from HyClone (Logan, UT, USA). The 100× nonessential amino acids, 100× penicillin and streptomycin, 0.25% trypsin with ethylene-diaminetetraacetic acid (EDTA), 1 M 4-(2-hydroxyethyl)-1-piperazi- neethanesulfonic acid (HEPES), and bovine serum albumin were purchased from Invitrogen Corp. (Carlsbad, CA, USA). Verapamil, MK571, indomethacin, benzbromarone, apigenin, sodium vanadate and cimetidine used in this study were obtained from Aladdin Industrial Inc. (Shanghai, China). Morroniside (purity > 98.0%) and loganin (internal standard (IS), purity > 98.0%) were obtained from the National Institute for the Control of Pharmaceutical and Biological Products (Beijing, China). All other reagents (typically of analytical grade or better) were used as received.

Phytochemical Glucuronidation Assay
